摘 要:Na_2SeO_3对人红细胞膜骨架具有稳定作用,但这种作用依赖于Na_2SeO_3的浓度。在低离子强度下,4℃透析人红细胞膜,实验组加入不同浓度的Na_2SeO_3,对照组不加Na_2SeO_3。结果表明,0.1—0.8ppm Na_2SeO_3的存在比对照组具有较高的Na^+,K^+-ATP酶活性、膜脂流动性。用N-[3-芘]-马来酰胺作探针,反映两者构象也有差异。如果在透析液中加入较高浓度的Na_2SeO_3(>1.0ppm)则会产生与低浓度相反的结果。人红细胞膜~31P-NMR的测试也表明,加入0.4ppm与4.0ppm Na_2SeO_3会产生不同的结果。与对照组相比较,低浓度使化学位移各向异性值(△σ)下降,而高浓度则使△σ增加。Effect of Na2SeO3 on the stabilization of human erythrocyte membrane is concentration-dependent. Human erythrocyte membranes were dialysed in low ionic strength medium at 4℃ in the absence (control) or presence of various concentrations of Na2SeO3. Results showed that a higher enzymatic activity of Na+,K+-ATPase and lipid fluidity were observed in the presence of 0.1-0.8ppm Na2SeO3 than that of the control. By using N-[3-pyenyl]-maleimide as probe, a conformation differencehas also been detected between the experimental and control samples.However, if the supplemented Na2SeO3 was in high concentration (>1ppm), results opposite to that with low concentration were obtained.An inverse result of 31P-NMR study of human erythrocyte membrane was also obtained with 0.4 PPm and 4.0 PPm Na2SeO3 supplementation. The chemical shift anisotropy(Δσ)increased in the order:0.4 Ppm Na2SeO3 <control(-Na2SeO3)<4.0ppm Na2Seo3.
