摘 要:目的建立茵陈蒿汤血清药理学研究方法。方法建立CCl4诱导的大鼠肝细胞损伤细胞模型,SD大鼠为含药血清供体,以含药血清对损伤肝细胞增殖(WST-8法)的影响为指标,采用正交试验法优选含药血清的制备方法,并确定体系中血清添加量。结果每天以成人等效剂量10倍的茵陈蒿汤水提醇沉液间隔8 h分2次给正常大鼠灌胃给药,连续3 d,末次给药后1 h采集血浆,体系中加入20%含药血清能明显提高损伤肝细胞的增殖能力。结论适宜条件下制备的茵陈蒿汤含药血清对CCl4诱导的肝细胞损伤具有明显保护作用,血清药理学研究方法可以用于该方剂的药效评价。OBJECTIVE To establish a seropharmacological method for Yinchenhao decoction. METHODS Model of CC14-induced injury of hepatocyte was established and rats were used as the donor to prepare Yinchenhao decoction containing serum. Hepatocyte proliferation and orthogonal experiment was carried out to screen the preparative method of drug-containing serum and the suitable concentration in culture system. RESULTS Normal rats as the donator of serum, ten times equivalent dose of the extract of Yinchenhao decoction for adults was used. The optimal preparative method of drug-contained serum was orally administration twice a day for 3 days, plasma was collected after 1 h of the last administration. Addition of 20% drug-contained serum to culture system could elevate hepatocyte proliferation significantly. CONCLUSION Yinchenhao decoction containing serum prepared by suitable method possesses an obvious protective effect on CC14-induced hepatocyte injury and seropharmacology can be applied to evaluate the pharmacological effect of Yinehenhao decoction.
