摘  要:目的考察白芍总苷(TGP)以及TGP和强的松(Pre)联合用药对豚鼠变应性接触性皮炎(ACD)的治疗作用。方法 90豚鼠随机分为8组:正常组,模型组,TGP(21、42、84 mg/kg)组,联合给药(TGP 42 mg/kg+Pre 1.4mg/kg)组和Pre(1.4、2.8 mg/kg)组。用二硝基氯苯诱导豚鼠制备ACD模型,各给药组均于实验第21天开始灌胃给药,正常组和模型组给予等体积的0.5%羧甲基纤维素钠,检测耳重量及耳厚度,采用放射免疫法测定豚鼠血清中白细胞介素(IL)-2和IL-4含量水平。结果 ACD豚鼠的耳片明显肿胀,并伴有大量的单核细胞浸润。TGP(42、84 mg/kg)以及联合给药组(TGP 42 mg/kg+Pre 1.4 mg/kg)能明显减轻ACD豚鼠炎症反应,改善其耳片病理,并降低血清中升高的IL-4水平,以及升高其降低的IL-2水平。此外联合给药组对豚鼠ACD的治疗效果明显优于TGP组以及Pre组。结论 TGP以及联合给药对豚鼠ACD有着良好的治疗作用,为ACD的临床治疗提供了新的依据。Objective To investigate the therapeutic effects of total glucosides of paeony (TGP) and the combina- tion of TGP and prednisone (Pre) on guinea pigs with allergic contact dermatitis (ACD). Methods Guinea pigs with ACD were induced by applying dinitrochlorobenzene (DNCB) to their skins repeatedly. The levels of intedeu- kin-2 (IL-2) and IL-4 in serums of Guinea pigs were measured by radioimmunoassay. Results The results showed that topical application of DNCB on the ear provoked inflammatory reactions with monocytes infiltration and ear swelling in Guinea pigs. The medication of TGP (42, 84 mg/kg) and the combination of TGP and Pre reduced in- flammatory reactions in ACD guinea pigs, ameliorated the histopathology of ear tissue, increased the levels of IL-2 and meanwhile, reduce those of IL-4 in the sera of ACD Guinea pigs. In addition, the therapeutic effects of TGP (42 mg/kg + Pre 1.4 mg/kg) on ACD mice were more effective than those of TGP (42 mg/kg) or Pre( 1.4 mg/ kg). Conclusion These results suggest that TGP and the combination of TGP and Pre both have good therapeutic effects on ACD, which can be clinically developed for the treatment of ACD.
