摘  要:目的研究雷帕霉素(RAP)对体外转化生长因子β1(TGF-β1)刺激的肾间质成纤维细胞NRK-49F的作用及其抗纤维化的作用机制。方法 TGF-β1 2ng/ml刺激肾间质成纤维细胞NRK-49F后,分为对照组(C组)及不同浓度RAP 20、40、60、80、100ng/ml处理组(分别为A1、A2、A3、A4、A5组);分别孵育24、48、72h;采用Western blot检测结缔组织生长因子(CTGF)、纤维连接蛋白(FN)表达的变化。结果与C组相比,不同浓度RAP均能降低CTGF和FN蛋白表达;其中,A2组抑制效应最为明显(P<0.05),24h时CTGF和FN蛋白表达分别下降45.11%和37.84%,48h时分别下降40.09%和45.68%,72h时分别下降49.62%和51.65%。结论 RAP可以抑制TGF-β1诱导的成纤维细胞CTGF和FN表达增加,提示RAP可能用于防治肾间质纤维化。Objective To investigate the effect and underlying mechanism of rapamycin(RAP) on the activation of transforming growth factor-β1 (TGF-β1)-induced renal interstitial fibroblast. Methods The renal interstitial fibroblast NRK-49F was stimulated by TGF-β1 2 ng/ml and then divided into 6 groups of C(normal controls), Al(treated with RAP 20 ng/ml), A2 (treated with RAP 40 ng/ml), A3 (treated with RAP 60 ng/ml), A4 ( treated with rapamycin 80 ng/ml) and A5 ( treated with rapamycin 100 ng/ml). After 24,48 and 72 h, the protein expressions of connective tissue growth factor(CTGF) and fibronectin(FN) were detected by Western blot. Results Compared with group C, different concentrations of rapamycin could inhibit the protein expressions of CTGF and FN, especially in group A2 (P〈0. 05). The protein expressions of CTGF and FN in group A2 were remarkably decreased by 45.11% and 37. 84% at 24 h,40. 09% and 45. 68% at 48 h and 49.62% and 51.65% at 72 h, respectively. Conclusion Rapamycin can inhibit TGF-β1-induced CTGF and FN increasement in the fibroblasts, which suggests that rapamycin may prevent renal interstitial fibrosis.
