摘 要:以陕西渭北旱塬最新审定的双低油菜品种合油杂2号以及亲本为试验材料,用SSR标记技术研究杂种与双亲之间的扩增谱带多态性以甄别真假杂种。结果发现,200对SSR引物先用亲本进行预选,有45对引物可以特异区分两亲本,进一步反复筛选,其中引物CB10524分别在杂交种和其双亲之间存在扩增条带的多态性,表现为杂交种条带均为父母本的互补型,很适合做杂交种纯度鉴定。用该引物对合油杂2号杂交种进行SSR纯度鉴定,所测纯度分别为93%,与田间实测纯度94.27%非常接近,表明开发的SSR标记技术在合油杂2号油菜杂交种子纯度室内快速检测中具有广阔的应用前景。In order to evaluate the purity of hybrid seeds,Brassica napus cultivar Heyouza No.2,as the newly registered and first double-low rapeseed variety in Weibei highland in Shaanxi province,and their parental inbred lines were used to study the polymorphorism of amplifiable spectral bands by 200 SSR primer pairs.The results showed that 45 primer pairs can distinguish two parental inbred lines specifically.Further study,one primer pair(CB10524) was found to be polymorphic in the hybrid seeds and their parents and the polymorphism was presented in the form that all the bands amplified from the hybrid seeds were complementary to those from their parents,which was suitable for identification of seed purity.The purity of Heyouza No.2 seeds were tested with the primer pair,and the seed purity was 93%,which was close to the result of 94.27% identified in the field.It indicated that the SSR marker would have a wide range of applications in rapid lab test of seed purities of B.napus hybrid cultivar Heyouza No.2.
