摘  要:[目的]探讨低强度微波辐射对盐酸阿霉素(DOX)致人早幼粒白血病HL-60细胞损伤的影响。[方法]将HL-60细胞随机分为对照组、单纯微波组、单纯DOX组和联合组(微波+DOX)。单纯微波组和联合组都给予12μW/cm2微波照射,1h/d,连续辐照3d,对照组和单纯DOX组置于同一环境,但不给予电磁辐射。第4天分别给予单纯DOX组和联合组以浓度为0.125mg/L的DOX。用钙离子依赖性磷脂结合蛋白-异硫氰酸荧光素/碘化丙啶(Annexin V-FITC/PI)检测细胞凋亡率,流式细胞仪检测线粒体膜电位和胞内游离Ca2+浓度。[结果]与对照组相比,单纯DOX组凋亡率明显增加(P<0.05);线粒体膜电位明显下降(P<0.05);胞内游离Ca2+浓度明显升高(P<0.05)。与单纯DOX组相比,联合组凋亡率明显降低(P<0.05);线粒体膜电位明显升高(P<0.05);游离Ca2+浓度明显降低(P<0.05)。[结论]预先900MHz、12μW/cm2低强度微波照射能够明显减轻DOX引起的细胞损伤。[ Objective ] To explore the protective effects of low intensity microwave on doxorubicin hydrochloride (DOX) induced damages in HL-60 cells. [ Methods ] HL-60 cells were randomly divided into control, microwave alone, DOX alone, and combined exposure groups. The microwave alone and the combined exposure groups were exposed to 12 μW/cm^2 microwaves, 1 hour per day for 3 days. The cells in the control and the DOX alone groups were placed in the same condition, but without microwave radiation. The combined exposure and the DOX alone groups were subsequently exposed to DOX at the concentration of 0.125 mg/L on the 4th day. Apoptosis was detected with Annexin V-FiTC/PI double staining. Mitochondrial membrane potential (MMP) and intracellular Ca^2+ concentrations were determined by flow cytometry. [ Results ] Compared with the control group, the apoptosis rate and Ca^2+ concentration of the DOX alone group significantly increased (P 〈 0.05), while the MMP statistically decreased (P 〈 0.05). Compared with the DOX alone group, the apoptosis rate and Ca^2+ concentration of the combined exposure group significantly decreased (P 〈 0.05), while the MMP statistically increased (P 〈 0.05). [ Conclusion ] Under this experimental condition, pre-exposure with 12μW/cm^2 900MHz microwaves could significantly alleviate D OX-induced damages to HL-60 cells.
