摘 要:目的:观察平喘Ⅰ号对呼吸道合胞病毒(RSV)感染引起BALB/c小鼠哮喘模型肺组织中核转录因子-κB(NF-κB)、转化生长因子-β(TGF-β)含量的影响,探讨其治疗哮喘发作的作用机制。方法:72只BALB/c小鼠随机分成6组,随机分为正常对照组、模型对照组、地塞米松组、平喘Ⅰ号低、中、高剂量组,每组12只。予RSV致敏,除正常对照组和模型对照组用等量生理盐水外,其余各组用相应药物干预。末次激发48h后取肺组织病理切片采用免疫组化法测定各组NF-κB和TGF-β含量。结果:与正常对照组比较,哮喘模型组小鼠肺组织中NF-κB、TGF-β含量明显增高(P<0.01)。与模型对照组比较,平喘Ⅰ号各剂量组和地塞米松组均可降低哮喘小鼠肺组织中NF-κB和TGF-β水平(P<0.01或P<0.05),其中平喘Ⅰ号高剂量组与地塞米松组水平接近。结论:平喘Ⅰ号能降低哮喘小鼠肺组织中NF-κB、TGF-β含量,并具有量效关系。由此推测,通过降低肺组织中NF-κB和TGF-β水平,从而减轻气道炎症反应和气道组织的重构,是平喘Ⅰ号治疗哮喘的作用机制之一。支气管哮喘(简称哮喘)是由多种细胞(如嗜酸性粒细胞、肥大细胞、淋巴细胞等)和细胞组分共同参与的气道慢性炎症性疾病。本病反复发作,可产生气道不可逆性狭窄和气道重塑,致使病情缠绵难愈。平喘Ⅰ号是治疗哮喘急性发作的经验方,经过长期的临床治疗和观察,证实对小儿哮喘发作有良好的治疗作用。本研究的主要目的是观察哮喘小鼠肺组织中核转录因子-κB(NF-κB)、转化生长因子-β(TGF-β)的含量变化,探讨平喘Ⅰ号治疗哮喘的可能作用机理。Abstract:Objective:To observe the influence of PingchuanⅠ on Lung organization Nucleus duplication factor - KB (NF- KB)and Transformation growth factor- β( TGF- β) levels of BALB/c mice asthma model infected by respiratory syncytial virus (RSV), and to explore the therapeutic mechanism of asthma attacks. Methods:72 BALB/c mice were ran- domly divided into six groups, which were normal control group, model control group, dexamethasone group, Pingchuan I low, middle and high -dose group, 12 rats in each group. Sensitized to the RSV,the normal control group and model con- trol group received normal saline, all the other groups with the corresponding drug intervention. 48h after the last chal- lenge, SABC was used to detect the NF - KB and the TGF - β levels in each group. Results : Compared with normal con- trol ,NF- KB and TGF- β levels in asthma model groups were significantly higher( P 〈 0.01 ). With the model control group, Pingchuan Ⅰ each dose group and dexamethasone group can reduce NF - KB and TGF - β levels in asthma mice (P 〈0.01 or P 〈 0.05 ),the levels in the high dose group were nearly equal to the levels in dexamethasone group. Conclu- sion : The Pingchuan Ⅰ can reduce NF - KB and TGF- β levels, and has dose - effect relationship. The results suggestedthat, by lowering the NF - KB and TGF -β levels, thereby reducing airway inflammation and airway remodeling organiza- tion are the mechanism of Pingchuan Ⅰ on treating asthma.
