摘  要:目的:采用经典的CCL4复合因素建立肝纤维化大鼠模型,探究健肝颗粒抗肝纤维化的作用机制。方法:100只雄性SD大鼠随机分组后,以CCL4腹腔内注射的方法建立大鼠肝纤维化模型,造模后施以由低、中、高剂量健肝颗粒,以秋水仙碱为阳性对照。比色法检测血清活性氧(ROS)水平;硫代巴比妥酸(TBA)法检测肝组织中匀浆中丙二醛(MDA)含量。结果:模型组大鼠血清ROS水平明显升高,健肝颗粒可显著降低ROS水平,尤其以低剂量组效果明显(P<0.05);健肝颗粒组大鼠肝组织中MDA含量明显低于模型组(P<0.01)。结论:健肝颗粒可抑制肝脏脂质过氧化物的形成,可能是其抗肝纤维化的作用机制之一。Objective : To investigate the mechanism underlying the possible anti - fibrotic effects of Jiangan granules on rats with carbon tetrachloride(CC14) induced liver fibrosis. Method~:One hundred male SD rats with liver fibrosis induced by intraperitoneal injection of CC14 were randomized to receive a low, intermediate, or high dose of Jiangan granules or the positive control colchicine. Serum reactive oxygen species(ROS) were measured colorimetrically. The levels of malondial- dehyde(MI)A) in liver tissue homogenates were assessed by the thiobarbituric acid (TBA)assay. Results:Rats receiving colchicine had significantly increased levels of ROS. In contrast, Jiangan granules was associated with a significant de- crease in ROS, especially the low dose (P 〈 0.05 ). Compared with the controls, rats given Jiangan granules had signifi- cantly lower levels of MDA( P 〈 0.01 ). Conclusions : Jiangan granules appear to gain an anti - fibrotic effect by inhibiting the production of lipid peroxides in the liver.
