摘 要:目的:观察创伤性休克大鼠防御素-5(RD-5)mRNA变化及大黄的影响作用。方法:健康SD大鼠56只,分为正常对照组(8只)、创伤性休克组(24只)以及大黄干预组(24只)。观察回肠RD-5m RNA表达,潘氏细胞形态学及大黄对其影响。结果:创伤性休克组和大黄干预组RD-5 mRNA表达量在伤后8h显著升高,伤后24h降低,但仍高于正常水平,而伤后72h降至正常水平以下;组间比较显示,休克组和大黄干预组伤后8h和24h值差异无显著性,而72h休克组显著低于大黄干预组;两组回肠潘氏细胞与对照组比较无明显形态学改变。结论:创伤性休克早期RD-5表达增强可能是机体的一种保护反应。大黄对创伤性休克人鼠早期RD-5的表达无明显影响作用,72h后能增强RD-5表达作用,对机体胃肠道有保护作用。Objective : To investigate the change of defensin 5 mRNA in traumatic shock rats and the effects of Rhu barb. Methods :56 healthy SD rats were divided randomly into control group (n = 8), traumatic shock group (n = 24) and Rhubarb intervention group ( n = 24). Observe the expression of RD 5 mRNA, morphology of Paneth cells and the in fluence of rhubarb. Results : The expression of RD 5 mRNA both in traumatic shock group and rhubarb intervention group had a significant rise since injured 8 hours and 24 hours later. The expression of RD 5 mRNA after 24 hours later was lower than8 hours later, but still higher than normal level, then lower normal levels after 72 hours. Comparison a mong groups showed that there was no significant difference between shock group and the rhubarb intervention group after injured 8 hours and 24 hours, but the shock group significantly lower than the rhubarb intervention group 72 hours later; Paneth ceils of the shock group and the rhubarb intervention group and control group had no significant morphology change. Conclusion:The increased expression of RD 5 in the early stage after the traumatic shock might be a host pro tective response. There is no obvious effects of rhubarb for the early expression of RD 5 in traumatic shock rats, but it enhances the expression of RD 5 after 72 hours and might protect gastrointestinal dysfunction.
