摘 要:目的 比较荧光定量 PCR与RT-PCR(定性)检测不同检材中的 HCV-RNA,评价荧光定量PCR技术测定HCV-RNA水平的价值。方法 采用荧光定量PCR及 RT-PCR(定性)技术同时检测了71例血液透析患者的血清、血浆、淋巴细胞、尿液标本中 HCV-RNA。结果 血清、淋巴细胞、尿液标本中 HCV-RNA的荧光定量 PCR检出率(46.38%、85.51%、80.00%),分别高于相应标本中HCV-RNA的RT-PCR(定性)阳性率(24.64、28.99%、20.00%),有统计学意义(P<0.05)。血清、淋巴细胞中同时检出HCV-RNA的一致率,荧光定量PCR法(39.13%)显著高于RT-PCR定性法(8.70%)(P<0.001)。HCV-RNA的RT-PCR定性阳性率不随 RNA定量水平均高而增高。结论与 RT-PCR(定性)相比,荧光定量PCR技术检测Objective The aim of the study is to compare amplisensor quantitative PCR with RT-PCR (qualitative) in detecting HCV-RNA of different specimen and to evaluate the technique of amplisensor quantitative PCR. Methods Serum, urine and lymphocyte of 71 hemodialysis patients were tested for hepatitis C viral RNA by amplisensor quantitative PCR and RT-PCR (qualitative). Results Positive rate of HCV-RNA in seurm, lymphocyte, urine by amplisensor quantitative PCR (46.38%. 85.51%, 80.00%)was signdicanily higher than that by RT-PCR respectively (24.64%, 28.99%, 20.00%)(P<0.05).The rate of simultaneously positive HCV-RNA in serum and lymphocyte was significantly higher by amplisensor quantitative PCR (39.13%) than by RT-PCR (8.70%) (P<0.001). There was no relationship between the positive rate of HCV-RNA by RT- PCR and the quantilative level. Conclusion Compared with RT-PCR, amplisensor quantitative PCR technique has higher sen- sitivity in detecting HCV-RNA.
