摘 要:目的:考察菩人丹对2型糖尿病大鼠胰岛素敏感性和胰岛β细胞功能的影响。方法:采用脂肪乳灌胃结合小剂量链脲佐菌素尾静脉快速注射法复制T2DM大鼠模型,菩人丹干预治疗4周后进行口服糖耐量试验,应用HOMA-IR和ISI评价胰岛素敏感性,HOMA-β、ΔI30/ΔG30、AUCI评价胰岛β细胞分泌功能,DI评价葡萄糖利用率。结果:菩人丹干预后HOMA-IR明显降低,ISI、HOMA-β、ΔI30/ΔG30及DI均显著升高。结论:菩人丹可以有效减轻T2DM大鼠的胰岛素抵抗,提高胰岛素降糖水平,改善胰岛素的早时相分泌。Objective To investigate the effect of PRD on insulin sensitivity and islet β cell function in T2DM rats. Methods The rats model of type 2 diabetes mellitus were established by lipid emulsion gavage combined with small dose of streptozocin (STZ, 30 mg/kg) tail vein injection. The T2DM rats were treated with PRD. Oral glucose tolerance test (OGTT) was applied after the treatment for 4 weeks. Insulin sensitivity was determined by HOMA-IR and ISI. Islet β cell function was estimated by HOMA-β, △I30/△G30 and AUCI. Glucose utilization was evaluated by DI. R.esuhs Compared with the T2DM rats, HOMA-IR of PRD treated rats was significantly reduced, while ISI, HOMA-β, △I30/△G30 and DI were all increased significantly, which was similar to the effect of positive control drugs. But there is no significant difference of AUCI between treatment groups and T2DM group. Conclusion PRD can reduce insulin resistance in T2DM rats, increase insulin sensitivity, elevate level of glucose utilization, and improve the early phase insulin secretion, but has no significant effect on the second phase insulin release.
