摘 要:目的通过99mTc-MIBI显像以明确冰片对大鼠脑内、脑外恶性肿瘤细胞P-gp的功能是否有影响。方法取Wistar大鼠,制成皮下和脑内C6脑胶质瘤(C6)和耐药C6脑胶质瘤(C6R)肿瘤模型,分成对照组、冰片单次应用组和冰片连续应用组,进行躯体99mTc-MIBISPECT显像和脑部99mTc-MIBI放射自显影,利用感兴趣区(ROI)技术分别计算C6和C6R的肿瘤/肌肉放射性比值和肿瘤/脑皮质比值(T/NT),并把C6的T/NT与C6R的T/NT相除得出两者间的比值(TC6/TC6R),对各组显像结果进行比较。结果 1、C6R皮下肿瘤、脑内肿瘤摄取99mTc-MIBI明显低于C6皮下肿瘤、脑内肿瘤;2、冰片单次应用后,与对照组比较,C6皮下肿瘤和脑内肿瘤摄取99mTc-MIBI无明显变化,而C6R皮下肿瘤和脑内肿瘤摄取99mTc-MIBI明显增高,同时皮下肿瘤和脑内肿瘤的TC6/TC6R均明显减低;3、冰片连续应用后,与对照组比较,C6皮下肿瘤摄取99mTc-MIBI无明显变化,而C6R皮下肿瘤、C6脑内肿瘤、C6R脑内肿瘤摄取99mTc-MIBI均明显增高,且皮下肿瘤TC6/TC6R有明显降低,而脑内肿瘤TC6/TC6R虽有所降低,但无统计学差异。4、冰片连续应用后,对大鼠肿瘤细胞摄取99mTc-MIBI的影响程度与冰片单次应用相同。结论冰片单次应用和连续应用对脑内、外耐药恶性肿瘤P-糖蛋白的功能有一定的抑制作用,两者影响的幅度相似。Objective To study the influence of borneol on P-glycoprotein of malignant tumor cell u- sing 99mTc-MIBI imaging. Methods Tumor bearing rat model was established by implanting rat C6 glioma ceil (C6) and multidrug resistance rat C6 glioma celI(C6R) into the left and right thigh muscle and into the left- and fight-basal ganglia. The rats were randomized into control group, borneol once treating group and bomeol continually treating group. The 99mTc-MIBI imaging was performed in the rats of control group, and in the rats after bomeol treating. Tumor-to-nontarget (T/NT) ratio was calculated, then T/NT of C6 and T/NT of C6R ra- tio was calculated(TC6/TC6R ratio). The degree of the 99mTc-MIBI uptake and the P-gp function were com- pared in the rats of different groups. Results 1 Accumulation of 99mTc-MIBI in C6R tumor was lower than in C6 tumor. 2 In bomeol once treating group, accumulation of 99mTc-MIBI in C6 thigh tumor and C6 brain tumor were same as in the control group, and accumulation of 99mTc-MIBI in C6R thigh tumor and C6R brain tumor were higher than in the control group. TC6/TC6R ratio in thigh and brain tumor in borneol once treating group was lower than in the control group. 3 In borneol continually treating group, accumulation of 99mTc-MIBI in C6 thigh tumor was same as in the control group, and accumulation of 99mTc-MIBI in C6R thigh tumor, C6 and C6R brain tumor were higher than in the control group. TC6/TC6R ratio in thigh tumor in borneol continually treating group was lower than in the control group, but TC6/TC6R ratio in brain tumor in bomeol continually treating group was lsame as in the control group. 4 The changing degree of 99mTc-MIBI uptake in the rats of borneol continually treating group was same as in borneol once treating group. Conclusion Borneol treating could increase uptake of 99mTc-MIBI in multidrug resistance rat tumor, and it implied that borneol was one of inhibitors of P-gp function.
