活血止痛膏对兔骨骼肌急性钝挫伤后Ⅱb型MHC、Ⅰ型及Ⅲ型胶原mRNA表达的影响

摘　　要：目的:观察活血止痛膏剂对兔骨骼肌钝挫伤后内源性IIb型肌球蛋白重链(MHC)、Ⅰ型及III型胶原mRNA表达的影响。方法:63只大白兔随机分为活血止痛膏高、中、低剂量组与青鹏膏对照组、外伤对照组,每组12只。另取3只为正常对照组。各实验组采用"重物自由落体打击法"制作大白兔骨骼肌急性钝挫伤模型。受伤第1天即于伤处贴敷相应药物。各组伤后3、10、20、27天各处死大白兔3只,取腓肠肌标本,共6个。RT-PCR技术检测内源性Ⅱb型MHC、Ⅰ型及Ⅲ型胶原mRNA。结果:①伤后第3、10、20天活血止痛膏高、中剂量组和青鹏膏组内源性Ⅱb型MHCmRNA表达量高于外伤组和低剂量组(P<0.05);第10、20天活血止痛膏高剂量组内源性IIb型MHCmRNA表达高于中剂量组(P<0.05);②内源性I型胶原mRNA表达量:第3天各组比较无统计学意义(P>0.05);第10、20天活血止痛膏高、中剂量组和青鹏膏组表达量低于外伤组、低剂量组(P<0.05);第10、20天中剂量组高于高剂量组(P<0.05)。③内源性Ⅲ型胶原mRNA表达量:第3、10、20天活血止痛膏高、中剂量组和青鹏膏组内源性Ⅲ型胶原mRNA表达低于外伤组和低剂量组(P<0.05);第3、10、20天高剂量组和青鹏膏组表达量明显少于中剂量组(P<0.05)。三个检测指标各个时间点青鹏膏组和高剂量组比较,差异无统计学意义(P>0.05)。结论:活血止痛膏可有效促进骨骼肌急性钝挫伤后Ⅱb型MHCmRNA表达,减少Ⅰ、Ⅲ型胶原mRNA表达,其作用与青鹏膏相当。ⅢⅢ Objective： To investigate the effect of Huoxuezhitong cream on the expression of endogenous type lib MHC mRNA, and I and Ⅲ collagen mRNA after muscle contusion in rabbits. Methods： A total of 63 rabbits were randomly divided into Huoxuezhitong cream high dose（n=12）, middle dose（n=12）, and low dose groups（n= 12）, Qingpeng ointment group（n=12）, trauma control group（n=12）, and blank control group（n=3）. The model of acute skeletal muscle contusion was established by using the method of free-falling weights in other 5 groups ex- cept the blank one. Each group was given the corresponding intervention treatment on day 1 after contusion. Rab- bits were killed on day 3,10,20,and 27 after contusion for harvesting specimens of gastrocnemius muscle to de- tect the expression of endogenous type Ⅱb MHC mRNA, and I and m collagen mRNA, 3 rabbits at each time point in each group. Results： As compared to the trauma and blank control groups, the high dose and middle dose groups of Huoxuezhitong cream had elevated expression of endogenous type H b MHC mRNA on day 3,10 and 20（P〈0.05） and the high dose and middle dose groups of Huoxuezhitong cream had higher level of endoge- nous type Ⅱb MHC mRNA than that in the low dose group of Huoxuezhitong cream on day 10 and 20（P〈 0.05）. The expression of endogenous type I collagen mRNA in each group was not different on day 3（P〉0.05）, but lower in the high dose and middle dose groups of Huoxuezhitong cream and Qingpeng ointment group on day 10 and 20 than that in the trauma control group and low dose group of Qingpeng ointment group（P〈0.05）; and the level was lower in the high dose group of Huoxuezhitong cream and Qingpeng ointment group（P〈0.05） .On day 3,10 and 20, the expression of endogenous type Ⅲ collagen mRNA in the high dose and middle dose groups and Qingpeng Ointment group was lower than that in trauma control group and low dose group （P〈0.05）, and lower in the high dose group and Qingpeng Ointment group
