新疆维吾尔族妊娠期高血压疾病胎盘中MMP-9、TIMP-1的表达及意义

摘　　要：目的探讨维吾尔族妊娠期高血压疾病(pregnancy induced hypertension,PIH)胎盘中MMP-9、TIMP-1的表达及与PIH的相关性。方法收集新疆自治区人民医院产科2009年12月～2010年8月住院分娩的维吾尔族患者胎盘组织标本90例,其中子痫前期重度组30例,子痫前期轻度组30例,正常组30例。结果 (1)胎盘组织中MMP-9与TIMP-1分布一致,主要分布在滋养叶细胞、血管内皮细胞及绒毛间质细胞的胞质中。重度组胎盘组织中MMP-9阳性颗粒分布明显减少。MMP-9在三组比较中,差异有显著性;(2)重度组与正常组、轻度组两两相比差异均有显著性,随着PIH病情的加重,MMP-9的阳性表达逐渐降低;(3)TIMP-1在各组中的表达,差异无显著性;(4)重度组MMP-9/TIMP-1阳性表达比低于正常妊娠组及轻度组,而且随着病情的加重,二者阳性表达比逐渐变小。结论 (1)PIH患者胎盘组织中MMP-9的阳性表达水平随着PIH病情加重,明显减少;(2)PIH患者胎盘组织中TIMP-1在各组中变化不大,与PIH病情无明显相关性.Purpose To detect the MMP-9 and TIMP-1 expression in Uighur pregnancy-induced hypertension（PIH） placenta and to analyze their relevance to PIH.Methods Placental tissues were collected in 90 cases of Uighur pregnancy woman in Xinjiang Autonomous Region People＇s Hospital during Dec.2008-Sept.2009,including 30 patients with severe pre-eclampsia,30 mild pre-eclampsia and,30 normal pregnancies.Results Placental distribution of MMP-9 and TIMP-1 expression was identical,mainly in the cytoplasm of trophoblastic cells,vascular endothelial cells and villous stromal cells.Positive staining of MMP-9 in the severe pre-eclampsia groups decreased.The MMP-9 expression in the severe pre-eclampsia was also significantly lower than that in the other groups（P0.000 1）.Among severe pre-eclampsia groups,normal group and mild group,there were statistically significant differences in the expression of MMP-9.The positive rates of MMP-9 become lower with severity state of illness.There was no significant difference in TIMP-1 expression among all the groups.The positive rates of MMP-9/TIMP-1 in the severe pre-eclampsia groups were lower than that of control groups and mild groups.The positive rates of MMP-9/TIMP-1 became lower with severity of illness.Conclusion Expression of MMP-9 in the PIH becomes significantly lower with severity of illness.There is no significant difference in TIMP-1 expression among all the groups,which is also not correlated with PIH.
