摘 要:目的观察大鼠心脏移植急性排斥反应中的移植心脏的存活和心肌细胞的凋亡情况及落新妇甙对其影响。方法进行36对SD-Wistar大鼠间的颈部心脏移植,术后将Wistar大鼠分为两组(各18只),落新妇甙组给予落新妇甙,对照组给予生理盐水。分别于术后第3、5和7天切取移植心脏,用苏木素-伊红(HE)染色和原位末端标记(TUNEL)技术检测移植心脏切片,进行排斥反应的病理分级并计算凋亡指数(AI),比较落新妇甙组和对照组间的差异。结果 (1)移植心脏的病理分级:移植后第3、5和7天对照组分别为0.77±0.31、1.99±0.62和3.44±0.54;落新妇甙组分别为0.37±0.42、0.88±0.20和1.04±0.33;两组比较差异均有统计学意义(均为P<0.01)。(2)细胞凋亡主要发生于心肌细胞;落新妇甙能明显减少心肌细胞的凋亡,减轻移植心脏的组织损伤。移植后第3、5和7天对照组AI分别为4.03±1.32、8.52±1.97和9.73±6.21;落新妇甙组分别为0.87±1.11、2.53±2.49和4.71±2.99;两组比较差异均有统计学意义(均为P<0.01)。结论在大鼠心脏移植模型中,落新妇甙能明显抑制心肌细胞凋亡,减轻移植心脏的急性排斥反应。Objective To investigate the effects of Astilbin on apoptosis during acute rejection of cardiac allograft in rats. Methods Thirty-six Wistar rats underwent heterotopic heart transplantation in the cervical portion from disparate SD rats and were equally divided into Astilbin group(treated with Astilbin) and control group(treated with saline).The cardiac grafts were harvested at 3,5 or 7 days after transplantation.The cardiac allograft sections were subjected to the HE staining and in situ terminal deoxynucleotidyl transferas-mediated deoxyuridine triphosphate nick end labeling(TUNEL) for histopathological rejection grade and apoptotic index(AI). Results Apoptosis mainly occurred in cardiac myocytes throughout allograft rejection.Astilbin could significantly reduce myocyte apoptosis and abate the damage to the graft tissue.The histopathological rejection grade at 3,5 or 7 days post-transplantation were(0.77±0.31),(1.99±0.62),and(3.44±0.54) in control group and(0.37±0.42),(0.88±0.20),(1.04±0.33) in Astilbin group(all P0.01).The AI at 3,5 or 7 days post-transplantation were(4.03±1.32),(8.52±1.97) and(9.73±6.21) in control group and(0.87±1.11),(2.53±2.49) and(4.71±2.99) in Astilbin group(all P0.01). Conclusions Astilbin can apparently inhibit the apoptosis of cardiac myocytes,which may be an important way of its immunosuppressive actions in acute cardiac allograft rejection in rats.
