摘 要:目的研究非诺贝特对高脂模型大鼠肝脏和主动脉中过氧化物酶增殖物活化受体γ(PPARγ),肝X受体(LXRα)和ABCA1 mRNA及其蛋白表达的影响。方法将30只雄性健康Wistar大鼠,随机分为正常对照组、高脂模型组、非诺贝特组,应用RT-PCR方法测量各组肝脏和血管中PPARγmRNA,LXRα和ABCA1 mRNA的表达,应用Western印迹方法测量肝脏和血管中PPARγ,LXRα和ABCA1蛋白的表达。结果高脂模型组肝脏、主动脉中PPARγ,LXRα和ABCA1 mRNA及蛋白表达水平与正常对照组相比升高,但差异无统计学意义(P>0.05)。非诺贝特药物干预后大鼠肝脏、主动脉的PPAR-γmRNA与模型组比较虽有升高,但差异无统计学意义(P>0.05);LXRα和ABCA1 mRNA与模型组比较差异有统计学意义(P<0.05);它们蛋白表达变化趋势与mRNA的表达均趋势一致。结论非诺贝特不仅降低血脂,而且能够升高LXRα的表达和促进ABCA1的释放,使ABCA1的含量升高,促进胆固醇的逆转运,发挥抗动脉粥样硬化的作用。Objective To explore effects of fenofibrate on the expressions of PPAR-r, LXRcαand ABCA1 mRNA in rats model with hyperlipaemia, and investigate the mechanism of regulating ATP binding cassette transporter A1. Methods 30 rats were randomly divided into control, model and fenofibrate treated groups. Expressions of PPAR-r, LXRα and ABCA1 mRNA were detected by RTPCR. PPAR-r, LXRα and ABCA1 mRNA and protein expressions in the liver and aorta were determined by RT-PCR and Western blot. Results Protein and mRNA expressions of PPARr, LXRα and ABCA1 in liver and aorta had no significant difference between control and hyperlipaemia group ( P 〉 0.05 ). The expressions of PPAR-r mRNA in liver and aorta in fenofibrate group were significantly higher than those of hyperli- paemia group, had no significant difference between fenofibrate group and hyperlipaemia group (P 〉 O. 05 ). The expressions of LXRα and ABCA1 mRNA in liver and aorta in fenofibrate group Were significantly higher than those of hyperlipaemia group, had significant difference between fenofibrate and hyperlipaemia groups (P 〈 0. 05 ). The changing trends of expression of PPAR-r, LXRα and ABCA1 protein and mRNA were consistent trend. Conclusions Fenofibrate could decrease blood lipids elevate the expressions of LXRα and ABCA1 in a non- dependent manner of PPAR-w.
