摘  要:目的 Torque teno病毒(Torque teno virus,TTV)是近年来才发现的一种单股负链DNA病毒。猪Torque teno病毒(Torque teno sus virus,TTsuV)在猪群中大量流行,被认为与猪的断奶后多系统衰竭综合征(post-weaning multi-systemic wastingsyndrome,PMWS)等疾病有关。本文旨在为广东TTsuV的流行病学提供数据。方法本文根据TTsuV的非编码区(UTR)合成引物,对来自广东2个猪场的14份猪血清进行了PCR检测。将2个猪场的TTsuV1和TTsuV2PCR产物各取1份,共4份PCR产物,进行克隆,测序并分析。结果 PCR共获得10份TTsuV1阳性(71%),8份TTsuV2阳性(57%),其中TTsuV1和TTsuV2双重阳性的为5份(36%)。PCR产物与参考毒株相应序列的分析证实,GDT1-1和GDT1-2的UTR片段均与TTsuV1参考毒株高度相似,GDT2-1和GDT2-2的UTR片段与TTsuV2参考毒株高度相似。结论广东至少存在两种血清型的TTsuV,在一些猪场有较高的检出率。TTsuV可能具有潜在的致病性,其公共卫生意义不容忽视。Human TT virus(TTV),a novel virus with negative single strand DNA,is ubiquitous and nonpathogenic.Viruses related to human TTV have since been identified in non-human primates,bovine,ovine,porcine,feline,and canine.Torque teno sus virus(TTsuV) was found widely prevalent in pig herds,and considered to involve with some diseases such as post-weaning multi-systemic wasting syndrome(PMWS).The objective of this study was to characterize TTsuV from pigs in Guangdong Province in South China.PCR primers were synthesized based on untranslated region(UTR) segment of TTsuV genome.PCR was performed by 50 cycles to detect 14 swine serum samples which came from two farms in Guangdong Province,following the method previously reported by Segalés et al in 1999.Two of the TTsuV1 PCR products,GDT1-1 and GDT1-2,and two of the TTsuV2 PCR products,GDT2-1 and GDT2-2,were cloned,sequenced and analyzed.PCR showed 71%(10/14) TTsuV1 positive,57.1%(8/14) TTsuV2 positive,and 36%(5/14) co-infectious.Sequence analysis showed that the UTR segments of sample GDT1-1 and GDT1-2,both of which were 305 bp,shared 90.2%-95.1% similarity with the TTsuV1 strains,and the UTR segments of sample GDT2-1 and GDT2-2,259 bp and 248 bp respectively,shared 67.3%-100% similarity with TTsuV2 strains.These outcomes conformed that at least two types of TTsuV existed in South China.Despite the putative harmlessness of TTsuV,the potential inoculum carried by pigs must be further evaluated as a sanitary threat.
