摘 要:利用单因素实验及响应面法优化确定胡芦巴种子多糖的提取工艺。通过单因素实验筛选出料液比、提取时间、提取温度三个主要因素,以胡芦巴种子多糖提取得率为响应值进行Box-Behnken中心组合试验设计,建立胡芦巴种子多糖提取得率的二次回归方程,得到优化组合条件。响应面法分析结果表明,当料液比为1∶28(g:mL),提取时间1.2 h,提取温度85℃时验证优化工艺得胡芦巴种子多糖最大提取得率19.89%,接近于模型预测值20.24%。Single-factor tests and response surface method (RSM) were used to optimize the extraction process of fenugreek seed polysaccharides (FSP). In order to obtaining the maximum of extraction yield of the polysaccharides, three main factors including extraction temperature, extraction times and ratio of raw material to water were selected with the Box-Behnken design and a quadric regression equation for predicting the yield of polysaccharides was established. The results showed that the optimum extraction conditions were as follows : extraction temperature 85 ℃, extraction times 1.2 h, and ratio of raw material to water 1: 28. Under the conditions,the optimum extracting yield of FSP was 19.89% ,which is well matched with the predictive extracting yield 20.24%.
