摘  要:目的探究中药独活对多发性硬化(MS)模型小鼠的治疗作用及机理。方法雌性C57BL/6小鼠,按文献方法制备实验性自身免疫性脑脊髓炎(EAE)模型。将造模小鼠随机分为正常,模型,独活低、中、高剂量,阳性对照药黄芩苷及醋酸泼尼松等7组。各组每日灌胃相应的药物,40天后分别进行EAE神经功能评分、淋巴细胞增殖能力检测、淋巴细胞分泌细胞因子含量测定。结果高剂量独活(36 mg/kg)能明显缓解EAE神经功能受损症状,使临床评分显著降低;能显著抑制淋巴细胞增殖;能使脾淋巴细胞分泌的IFN-γ浓度下降、IL-13浓度升高,上述结果与EAE模型组比较均差异显著(P<0.05,P<0.01)。结论独活能明显减轻EAE神经功能受损症状;显著抑制EAE小鼠脾淋巴细胞的增殖;下调Th1细胞因子IFN-γ、上调Th2细胞因子IL-13,恢复Th1/Th2的平衡,从而有效缓解MS/EAE。Objective To explore the therapeutic effects and immunological mechanisms of traditional Chinese medicine Duhuo on the mice model of multiple sclerosis. Methods Female C57BL/6 mice were used for the establishment of experimental allergic en- cephalomyelitis (EAE) model according to the reports, then randomly divided into seven groups. Drugs were administered p. o once a day for 40 days, and clinical scores, capacity of lymphocyte proliferation and cytokines levels secreted by lymphocyte were detected. Results Duhuo of high dose (36mg/kg) reduced clinical scores of EAE mice and inhibited the proliferation of lympho- cyte obviously (P 〈 0.01 vs EAE model group). It also decreased IFN -γ while increased IL- 13 secreted by lymphocyte (P 〈 0.01 vs EAE model group). Conclusion Duhuo can alleviate CNS damage in EAE, inhibit the proliferation of lymphocyte and re- store the balance of Thl/Th2, thereby alleviating MS/EAE.
