大鼠体内红景天苷浓度的测定及药代动力学研究

摘　　要：目的建立一种HPLC方法测定大鼠血浆和器官中红景天苷浓度,研究大鼠单剂量静脉注射红景天苷后器官分布情况及药代动力学模式。方法 HPLC色谱柱为Phenomenex C18(250 mm×4.6 mm id,5.0μm),流动相为甲醇-水(20∶80,V/V),流速为1.0ml/min,柱温为25℃,检测波长为278 nm。大鼠经股静脉注射单剂量红景天苷,于不同时间点采集血浆和器官样品,检测其中红景天苷的浓度。采用DAS软件对血药浓度-时间曲线进行统计矩拟合,得出系列药代动力学参数。结果 HPLC方法检测红景天苷的检测限为1.2 ng/ml,日内和日间精密度均小于8.2%,提取回收率大于76.4%。大鼠单剂量静脉注射红景天苷后,其药代动力学呈一房室模型特征。结论采用HPLC方法可精密快速检测出血浆及器官匀浆中红景天苷的浓度,大鼠单剂量静脉注射红景天苷的半衰期(t1/2)约为4 h,表观分布容积(Vd)为1.46 L/kg。Objective To develop an HPLC method for determination of salidroside in rat plasma and organs and to study on its organ distribution and pharmacokinetie profile after a single dose intravenous administration. Methods Analysis of salidroside in plasma and homogenate was carried out on a Phenomenex Cls （250 mm x 4.6 mmid, packed with 5.0μm C18 silica RP particle）. The mobile phase was consisted of methanol - water （ 20 ： 80 ） with the flow rate of 1.0 ml/min. The column temperature was main- tained at 25℃ and the detection wavelength was set at 278 nm. Rat plasma and organs were collected at different time points and the content of salidroside in them was detected after a single dose intravenous admininistration. The plasma concentration - time curve was fitted by DAS software. Results Detecting limit of the established HPLC method was 1.2 ng/ml. The variability values of intra - assay and inter - assay were both less than 8.2%. The extraction recovery was more than 76.4%. The pharmacokinetic profile showed one compartment model after one single dose intravenous injection. Conclusion The HPLC method is simple, accu- rate, rapid and applicable for pharmacokinetic study of salidroside in rats. The half life time and apparent volume of distribution of salidroside intravenous injection 4h and 1.46L/kg, respectively.
