摘 要:目的通过建立湿热证大鼠模型,观察大鼠血清IL-23和IL-17含量水平变化,探讨两者在湿热证发病中的作用和意义。方法将64只大鼠随机分为正常组、湿热模型组,通过高脂饮食、湿热环境、微生物感染三因素结合的方法制备湿热证大鼠模型。ELISA法检测造模后4,24,48,72 h两组大鼠血清IL-23和IL-17含量水平。结果模型组IL-23和IL-17的含量水平在各时相点均明显高于正常组(P<0.05);IL-23和IL-17均在造模后4h最高,24,48,72 h逐渐下降,48 h和72 h组均与4 h组比较有差异(P<0.05),72 h与24h比较亦有明显差异(P<0.05);IL-23和IL-17的表达水平呈正相关(P<0.01)。在轻度、中度、重度湿热证大鼠血清中,IL-23和IL-17的浓度水平均呈逐渐升高趋势,与湿热证的病情严重程度有正相关性(P<0.01)。结论 IL-23和IL-17参与了湿热证的发生、发展过程。两者的含量水平变化与湿热证的病情严重程度密切相关。IL-23与IL-17表达水平密切相关,在湿热证中IL-23可能通过促进IL-17表达水平增高而发挥其功能作用。Objective To observe the serum levels of IL - 23 and IL - 17 and its significance in syndrome of damp - heat . Meth- ods 64 rats were randomly divided into normal group and damp - heat model group. The damp - heat syndrome rat model was es- tablished by high -fat diet, hot and humid environment and microbial infeetion method. The levels of IL - 17 and IL -23 in serum of rats were tested by ELISA at 4h,12h,24h,48h. Results The levels of IL - 17 and IL -23 in rat model with syndrome of clamp -heat were significantly higher than those in normal group in different time( P 〈 0.05 ). The levels of IL -23 and IL - 17 were the highest at 4h after modeling, and were decreased gradually at 24h ,48h and 72h. The 48h and 72h groups of IL -23 and IL - 17 were significantly different in comparison with the 4h group( P 〈 0.05 ). 72 h group and 24h group also had distinct differences ( P 〈 0.05 ). Serum contents of IL - 23 was significantly positively correlated to IL - 17 in rat model with syndrome of damp - heat (P 〈 0.01 ). In mild, moderate and severe rat model with syndrome of damp - heat, the serum contents of IL - 23 and IL - 17 showed a gradual increasing trend with the clinical symptoms and serum eontents of the two were positively correlated to the severi- ty of syndrome of damp - heat significantly(P 〈 0.01 ). Conclusion IL - 23 and IL - 17 maybe participate in the pathogenesis of syndrome of damp - heat, and the two are significantly positively correlated to the severity of syndrome of damp - heat. Serum con- tent of IL - 23 is significantly positively correlated to IL - 17. The function of IL - 23 may be achived through affeeting the expres- sion of I1 - 17.
