摘  要:目的研究玻璃海鞘提取物分离组分的抗肿瘤活性。方法利用磺酰罗明B蛋白染色、吉姆萨与Hoechst染色、流式细胞术方法来观察相关指标。结果玻璃海鞘提取物分离组分对三种肿瘤细胞B16F0、Hela、HL-60有明显的抑制细胞增殖的作用,IC50分别为2.7,2.0,2.4μg/ml;在浓度为4.9μg/ml时,细胞体积变大,成梭形,出现树突状结构,有凋亡小体的出现;在一定浓度范围内,可以诱导B16F0细胞G2/M期阻滞。结论玻璃海鞘提取物分离组分具有显著的抗肿瘤活性,并诱导B16F0细胞G2/M期阻滞。Objective To investigate the antitumor activities of the purification component from Ciona intestinalis extractions. Methods The relevant indices were observed by protein sulforhodamine B coloration, Giemsa and Hoechst staining and flow cy- tometry methods. Results The component from Ciona intestinalis extractions had obvious proliferation inhibition on B16F0, Hela and HL-60 cells,and ICs0 was 2.7,2.0 and 2.4 wg/ml ,respectively. At a concentration of 4.9 p.g/ml,cell volume was in- creased into clostridial form, meanwhile, dendritic structure and apoptotic bodies emerged. It could induce GJM phase arrest on B16FO ceils in a certain range of concentration. Conclusion Purification component from Ciona intestinalis extractions possesses high antitumor activities,and it can induce G2/M phase arrest in B16F0 ceils.
