束缚-浸水应激对石榴皮多酚在大鼠主要器官内分布的影响

摘  要:目的观察束缚-浸水应激(RWIS)对石榴皮多酚在大鼠主要器官内分布的影响。方法利用FeCl3与多酚反应显色、与梯度多酚构成色阶,比色法检测多酚在器官内的含量。Wistar雄性大鼠250~300 g,灌胃给药,随机分为对照Ⅰ组(2 ml去离子水+应激,n=5);对照Ⅱ组(500 mg/ml石榴皮水提物、未应激)和实验Ⅰ、Ⅱ、Ⅲ(分别为5,50,500 mg/ml石榴皮水提物+应激)3个剂量组。给药1 h后行RWIS 3 h。过量麻醉处死动物后,心脏灌流除血,取脑、心、胃、肺、肝、肾、回肠、结肠匀浆,离心,取上清液与FeCl3反应。结果检测石榴皮多酚的最佳FeCl3浓度为9%。呈色反应的石榴皮水提物浓度为0.2~0.5 mg/ml。在大鼠的脑、心、肺、肝、肾上清液中未观察到呈色反应。实验Ⅲ组胃、结肠上清液与FeCl3反应阳性;而对照Ⅱ组的回肠、结肠也呈阳性,且高于实验Ⅲ组。结论 9%FeCl3可检测出0.2~0.5 mg/ml石榴皮水提物中的多酚。石榴皮多酚在胃肠壁中分布多于其它器官组织。RWIS使大鼠胃壁中石榴皮多酚含量增多,回肠和结肠壁内含量减少。Objective To observe the effect of restraint and water- immersion stress (RWIS) on the distribution of pomegranate peel polypbenol (PPP) in the major organs of rats. Methods The polyphenol content in rat organ was determined by the colorimet- ric method. Wistar male rats were randomly divided into control group I (2ml deionized water + RWIS, n = 5 ) , control group Ⅱ (500 mg/ml Aqueous extraction of pomegranate peel (AEPP) , without stress)and experimental groups Ⅰ , Ⅱ, Ⅲ ( 5,50.500 mg/ml AEPP + RWIS). 1 h after drug administration, the rats received RWIS for 3 hours, then the rats were killed by an over- dose of onesthesia and brain, heart, stomach, lung, liver, kidney, ileum and colon were harvested, homogenate , centrifuged, and the supeMatant was mixed with ferric chloride, color- reaction was observed. Results 9% ferric chloride was the best conatin for detection of PPP of AEPP. The response was obvious with AEPP 0.2 - 0.5 mg/ml. The supernate of brain, heart, lungs, liv- er, kidney had no color reaction. The supernates of stomach, colon were positive reaction with ferric chloride in experimental group Ⅲ; in the control group Ⅱ ileum, colon also showed positive reaction, which was higher than that in the experimental group Ⅲ. Conclusion 9% ferric chloride can sensitively detect content of PPP in 0.2-0.5 mg/ml AEPP. The PPP in gastrointestinal wall is distributed more than in other organs. RWIS is contribute to PPP enrichment at stomach wall, but reduce the PPP content at ile- um wall and colon wall.
