摘 要:目的探讨IFRD1表达下调后人肝癌细胞SMMC-7721基因表达谱的改变。方法采用脂质体lipo2000将构建的含目的基因shRNA的质粒转染入SMMC-7721细胞,并应用Affymetrix U133 plus 2.0 array基因芯片检测目的基因表达下调后,该细胞基因表达谱的变化。结果 RNA干扰抑制IFRD1基因表达后,基因芯片检测发现25476个基因中,表达量发生明显改变的有4221个(上下调大于等于1.5倍),上调的基因有1109个,下调的有3 112个;这些基因涉及到细胞周期、细胞凋亡、DNA复制等通路。结论 IFRD1基因表达下调后,人肝癌细胞SMMC-7721基因表达谱发生明显变化,与细胞生长增殖关系密切的细胞信号通路也发生了明显的改变。Objective To explore the change of gene expression profiles after IFRD1 gene down - regulation by RNA inference. Methods The alteration of gene expression profiles after IFRD1 gene silencing was investigated through Affymetrix HU133 plus 2. 0 gene chip,and bioinformatics was used to analyze data. Results Microarray assay revealed that expression of 4221 genes were obviously altered in response to IFRD1 gene silencing. Among them there were 1109 up -regulated genes and 3112 down -regula- ted genes. These genes were involved in cell cycle, apoptosis,, DNA replication, etc. Conclusion After the silence of IFRD1 gene, many genes have a significant differential expression. Some pathways related to cell growth and proliferation are also altered.
