摘 要:从长期受硝基苯严重污染场地中筛选出一株以硝基苯为唯一碳源和氮源的降解菌,命名为ZG。对ZG进行了16S rDNA和nbzA基因克隆与序列分析,并研究了该菌的降解特性。结果表明,ZG菌为革兰氏阴性细菌,初步鉴定该菌株为pseudomonas putida。ZG菌含有nbzA基因,并且该基因在质粒上,说明该菌降解硝基苯是通过半还原途径。ZG菌对硝基苯浓度、培养温度、pH值等外界环境因素具有一定的适用范围,但随着硝基苯浓度升高,硝基苯的降解速率和降解率表现出下降的趋势。在最佳降解条件下,即硝基苯浓度300 mg/L、温度20℃、pH为7,ZG菌对硝基苯的降解率达到99.98%。ZG菌可适用于不同的硝基苯污染场地。One degradation bacterium,using nitrobenzene as the sole carbon and nitrogen source,was screened from the long-term nitrobenzene-contaminated site and named as strain ZG.The strain ZG was cloned and sequenced through 16S rDNA and nbzA gene and the degradation characteristics were also studied.The results showed that the strain ZG was gram-negative bacterium and identified as pseudomonas putida initially.Strain ZG contained nbzA gene,which located in plasmid,illuminating that the pathway of strain ZG to degrade nitrobenzene was partial reduction.The strain ZG had a certain application scope to environmental factors such as nitrobenzene concentration,cultured temperature and pH value.However,the degradation speed and efficiency of nitrobenzene showed a downward trend with the rising of nitrobenzene concentration.Under the best condition,that was the nitrobenzene concentration of 300 mg / L,temperature of 20 ℃,pH=7,the nitrobenzene degradation rate by strain ZG was up to 99.98%.The strain ZG could be applicable to different nitrobenzene-contaminated sites.
