摘  要:目的研究珍珠菜抗肿瘤有效部位ZE4对肝癌Bel-7402细胞裸鼠移植瘤的抑制作用及可能机制。方法制备肝癌Bel-7402细胞裸鼠移植瘤模型,裸鼠分别ig给予ZE4 100,200和400 mg.kg-1每天1次,或ip给予5-FU 20 mg.kg-1,隔日1次,共21 d。移植肿瘤细胞后每3 d测量移植瘤体积并绘制生长曲线,末次给药后处死小鼠,剥离移植瘤组织称瘤质量并计算抑瘤率。应用Western印迹法检测移植瘤组织匀浆中凋亡相关蛋白Bax和Bcl-2蛋白表达,免疫组织化学法检测移植瘤组织中CD34表达,计数微血管密度(MVD)。结果 ZE4可抑制裸鼠移植瘤的生长,给药21 d后,ZE4 200 mg.kg-1组瘤质量为(1.5±0.5)g,较模型组(3.2±1.0)g明显减轻(P<0.01),抑瘤率达52.1%;移植瘤体积在给药14 d后为(0.83±0.27)cm3,与模型组(1.74±0.67)cm3相比,明显减小(P<0.05),给药21 d后为(1.40±0.42)cm3,比模型组(2.90±0.89)cm3相比,明显减小(P<0.05)。ZE4 200 mg.kg-1组Bax/Bcl-2比值(1.157±0.058)较模型组(0.253±0.008)显著升高(P<0.01),ZE4 100和400 mg.kg-1组无明显变化。ZE4 100,200和400 mg.kg-1组血管生成均明显减少,其中ZE4 200 mg.kg-1组MVD(13.5±1.0)较模型组(42.3±2.5)mm-2明显降低(P<0.01)。结论 ZE4对肝癌Bel-7402细胞裸鼠移植瘤具有明显抑制作用,可能与促进细胞凋亡和抑制血管生成有关。OBJECTIVE To study the inhibition of ZE4,the antitumor effective part from Lysimachia clethroides Duby,on Bel-7402 cell xenografted hepatoma and its possible mechanism.METHODS The sc transplanted tumor model of Bel-7402 cells in nude mice was established and then divided randomly into five groups,including model group and ZE4 100,200 and 400 mg·kg-1 and 5-fluorouracil(5-FU) 20 mg·kg-1 groups.The mice were ig given ZE4 once daily or ip given 5-FU every two days,for 21 d.The tumor volume was measured every three days after transplantation of Bel-7402 cells before the growth curve was made.All the mice were sacrificed after the last administration.The masse of the dissected tumor tissue were mearsured and the inhibitory rate was calculated.The expression of apoptotic protein Bax and Bcl-2 in transplanted tumor tissue was detected using Western blotting.CD34 expression in transplanted tumor tissue was detected with immunohistochemistry and microvessel density(MVD) was counted.RESULTS ZE4 could inhibit the growth of transplanted tumor in nude mice after administration for 21 d.The transplanted tumor mass in ZE4 200 mg·kg-1 group was(1.5±0.5)g,which significantly decreased(P0.01) compared with that of model group(3.2±1.0)g.The inhibitory rate was 52.1%.The transplanted tumor volume in ZE4 200 mg·kg-1 group was(0.83±0.27)cm3 after administration for 14 d,which was smaller than in model group(1.74±0.67)cm3(P0.05).After administration for 21 d,the transplanted tumor volume in ZE4 200 mg·kg-1 group(1.40±0.42)cm3 was also significantly smaller than that in model group(2.90±0.89)cm3(P0.05).Compared with model group(0.253±0.008),the ratio of Bax/Bcl-2(1.157±0.058) in ZE4 200 mg·kg-1 group significantly increased(P0.01),but there was no significant change in ZE4 100 and 400 mg·kg-1 groups.Immunohistochemistry analysis showed ZE4 100,200 and 400 mg·kg-1 could effectively reduce angiogenesis.ZE4 200 mg·kg-1 reduced MVD to 13.5±1.0 from model group(42.3±
