摘 要:以北京颐和园的古柿树当年新生的嫩芽切段为外植体进行组培快繁研究。结果表明,不定芽启动培养基为1/2MS+0.8 mg/L 6-BA+0.1 mg/L NAA+0.5g/L PVP。将无菌苗转接到最适分化培养基(1/2MS+5.0mg/L 6-BA+0.2mg/L NAA+0.5g/L PVP)上,组培苗分化效率较高,但小苗矮小。将分化较好的组培苗转接到壮苗培养基(1/2MS+2.0mg/L ZT+0.1mg/L IAA+0.5g/L PVP),待组培苗长至2~3cm转到生根培养基(1/2MS+0.6mg/L NAA+0.3mg/L IBA+0.5g/L PVP),暗培养9~10d后转接到生根培养基(1/2MS+0.5g/L PVP),生根率为30%。而将组培苗茎段预先用250mg/L的IBA或IAA浸泡2min,生根率可提高到40%。生根组培苗经炼苗后移栽成活率可达60%。An efficient and reproducible method for the regeneration of multiple shoots of Ancient Diospyros kaki Thunb from the Summer Palace has been developed in flasks containing primary shoots as the explants.The results showed that explants derived from in vitro grown seedlings were cultured on 1/2MS supplemented with 0.8 mg/L 6-benzylaminopurine(6-BA),0.1 mg/L 1-naphthlcetic acid(NAA),and 0.5 g/L polyvinylpyrrolidone(PVP).In addition,1/2 MS supplemented with 5.0 mg/L 6-BA and 0.2 mg/L NAA was found to be suitable for adventitious shoot induction and for the multiplication of individual shoots.However,the growth rate of the shoots was very low on the above medium.To mediate the growth and propagation of the shoots,selected shoots were transferred to the medium of 1/2MS+2.0 mg/L ZT+0.1 mg/L IAA+0.5 g/L PVP.In-vitro-produced shoots were rooted using a two-step method.Firstly,shoots were cultured on 1/2 MS with 0.6 mg/L NAA,0.3 mg/L IBA and 0.5 g/L PVP.Secondly,the shoots were transferred to 1/2 MS with 0.5 g/L PVP.The most effective first-step treatment was found to be dark training for 9~10 days,which initiated rooting at a frequency of 30%.If the shoots were immersed in 3-indolebutyric acid(IBA) or indole-3-acetic acid(IAA) at 250 mg/L for 2 min,and then transferred to 1/2 MS with 0.5 g/L PVP,the initiated rooting was at a frequency of 40%.Well-rooted plants were transferred to plastic cups containing vermiculite and perlite and successfully established in pots.This procedure is suitable for large-scale propagation for Ancient Diospyros kaki Thunb.
