摘 要:玻璃化现象是紫花苜蓿组培过程中常见的现象,由于其难以发育成苗或很难移栽成活而严重影响组织培养成功率。以和田苜蓿下胚轴为外植体,以MS为基本培养基,研究了在不同激素配比、蔗糖、琼脂浓度下和田苜蓿再生苗的玻璃化现象,并探索出改善玻璃化现象的措施。研究结果表明:在光照强度为1 000~1 200lx条件下,MS培养基中添加2.0mg/L 6-BA和0.5mg/L NAA,调整蔗糖浓度20~25g/L、琼脂浓度为7g/L时可有效降低和田苜蓿再生苗玻璃化现象。Vitrification is a common phenomenon for tissue culture of alfalfa.It is a serious problems which limits the success rate of plant tissue culture because they can not develop into normal plants or hardly survive when transferred from in vitro to greenhouse conditions.In the study,vitrification phenomenon of Hetian alfalfa in tissue culture was tested by change some factors(compounding proportions of different hormones,sugar concentration,agar concentration) as hypocotyls of alfalfa(Hetian) was used as explants and MS was the basic medium of tissue culture of hetian alfalfa.The results showed that the rate of vitrification can be reduced effectively on MS basal salt and vitamins contained 2.0 mg/L 6-BA and 0.5 mg/L NAA and 20~25 g/L sugar and solidified and 7g/L agar with 1 000~1 200 lx light intensity.
