消痰通腑方对结肠癌肝转移模型小鼠胰岛素生长因子蛋白表达的影响

摘　　要：目的从基因和蛋白水平研究消痰通腑方对结肠癌肝转移模型小鼠肝组织中胰岛素生长因子-I(IGF-Ⅰ)及其受体(IGF-ⅠR)、结合蛋白3(IGFBP-3)表达的影响,并探讨其机制。方法将造模成功的小鼠随机分为模型组、中药组,另设空白组,每组10只,中药组给予消痰通腑方灌胃,每日1次。待模型组小鼠成恶病质状态时,处死小鼠,取瘤组织及肝组织,观察各组荷瘤鼠肿瘤生长情况及抑瘤率、肝转移率及肝转移灶大小,肝质量及肝脏指数。并运用免疫组化法检测各组小鼠肝脏组织IGF-Ⅰ、IGF-ⅠR、IGFBP-3蛋白表达。结果模型组及中药组肝转移率均为100%,但中药组肝转移灶小于模型组(P<0.05);中药组小鼠瘤质量、肝质量、肝脏指数均低于模型组(P<0.05),与空白组比较,差异有统计学意义(P<0.05);中药组小鼠肝组织中IGF-Ⅰ、IGF-ⅠR蛋白表达均低于模型组(P<0.05),与空白组比较,差异无统计学意义(P>0.05)。中药组小鼠肝组织中IGFBP-3蛋白表达高于模型组(P>0.05),与空白组比较,差异无统计学意义(P>0.05)。结论消痰通腑方可以调节结肠癌肝转移组织中IGF-Ⅰ、IGF-ⅠR、IGFBP-3蛋白表达,这可能是其抗结肠癌肝转移的机制之一。Objective To observe the effects of Xiaotan Tongfu Recipe on IGF-Ⅰ/IGF-ⅠR protein expression in liver tissue of mice with tumor-burdened colon cancer and explore its mechanism on colon liver metastasis.Methods The successful model of mice were randomly divided into model group and Xiaotan Tongfu group,with another normal group as control,10 mice for each group.Xiaotan Tongfu group was given herbal medicine once per day.End of the experiment,the mice were killed and the tumor weight,tumor suppression rate,liver weight,liver index were observed.IGF-Ⅰ,IGF-ⅠR and IGFBP-3 of the liver were detected by immunohistochemistry.Results The tumor weight,liver weight and liver index of Xiaotan Tongfu group were significantly lower than that of the model group（P0.05）,compared with the normal group,it was no significant difference（P0.05）.IGF-Ⅰ and IGF-ⅠR protein of Xiaotan Tongfu group was lower than that of model group（P0.05）,and had no significant difference with the normal group（P0.05）.IGFBP-3 protein of Xiaotan Tongfu group was higher than that of Xiaotan Tongfu group（P0.05）,and had no significant difference with the normal group（P0.05）.Conclusion Xiaotan Tongfu Recipe could obviously inhibit the tumor weight of tumor-bearing mice,regulate the protein expression of IGF-Ⅰ,IGF-ⅠR and IGFBP-3 in liver tissue of mice with tumor-burdened colon cancer,which may be one of the mechanism of Xiaotan Tongfu Recipe anti liver metastasis of colon cancer.
