髓复康对大鼠脑缺血损伤区内勿动蛋白及其受体表达的影响

摘  要:目的观察益气活血补肾方髓复康对大鼠脑缺血损伤区勿动蛋白(Nogo)-A及其受体(NgR)表达的抑制作用。方法除正常组外,采用Koizumi法制作SD大鼠单侧大脑中动脉阻塞模型(MACO),并随机分为模型组、阳性对照组及髓复康大、中、小剂量组,各组在给药8、15、30 d 3个时间点取大鼠脑组织,采用免疫组化方法检测各组脑缺血损伤区Nogo-A及NgR表达量的差异。结果髓复康大、中、小剂量组Nogo-A、NgR表达减弱,与模型组比较差异有统计学意义(P<0.05,P<0.01)。结论髓复康可抑制脑缺血损伤区Nogo-A、NgR表达,改善脑缺血损伤区轴突再生微环境,这可能是其促进脑缺血损伤后轴突再生的机制之一。Objective To study the effect of Chinese herb Suifukang on the expression of Nogo protein-A and NgR in the brain of MCAO rats and explore its mechanism of promoting neurogenesis.Methods MACO model was made by Koizumi method.SD rats were randomized into normal group,control group,treatment of methylprednisolone group,treatment of Suifukang group in high-,medium-,low-dose.The brain tissue was taken at 8,15,30 d of drug administration.The expression of Nogo-A and NgR was detected by immunohistology method.Results The expression of Nogo-A and NgR in the injured brain were apparently inhibited by Suifukang(P0.01).Conclusion Suifukang can inhibit the expression of Nogo-A and NgR,which may be one of the mechanisms that Suifukang can promote the axon regeneration and the functional recovery after brain injury of cerebral ischemia.
