柴黄益肾颗粒对糖尿病肾病大鼠肝损伤保护作用的实验研究

摘　　要：目的:探讨柴黄益肾颗粒对实验性糖尿病肾病大鼠肝损伤的保护作用。方法:健康雄性Wistar大鼠112只,顺应性喂养3d后,随机分为假手术对照组12只,单侧肾切除组100只,造模成功后随机分为:模型组、柴黄益肾大、中、小剂量组、蒙诺组。观察柴黄益肾颗粒对大鼠肝重指数、血糖、血脂、24小时尿蛋白定量、肝组织糖原蓄积程度、肝组织Ⅰ、Ⅲ型胶原及肝组织病理的影响。结果:与模型组比较,柴黄益肾颗粒能够显著减低大鼠肝重指数(P<0.05),降低血清总胆固醇、甘油三酯以及尿液中24h尿蛋白定量(P<0.05);与蒙诺组比较,柴黄益肾颗粒能够显著减轻大鼠肝组织糖原蓄积程度、炎症活动度,同时柴黄益肾颗粒抑制了Ⅰ、Ⅲ型胶原在肝组织中的表达。结论:柴黄益肾颗粒对糖尿病肾病大鼠肝损伤具良好的保护作用。Objective： To investigate the effect of Chaihuangyishen Granule（CHYS） to liver histological injury in diabetic nephropathy rats which were induced by Streptozotocin（STZ）.Methods： After 3 days feed,Wistar rats were randomly divided into12 rats in sham control group and100 rats in uninephrectomy groups which were induced into diabetic modelby injection of 40mg/kg STZ.The diabetic rats were then randomly divided into model group,CHYS low dosage group,CHYS medial dosage group,CHYS high dosage group and Fosinopril group.The body weight,blood glucose,24h urineprotein of rats in six groups,were examined every four weeks.After 20 week treatment,all rats in the six groups were sacrificed.The level of serum TC and TG were examined.In order to examine the glycogen accumulation,the liver tissues were stained by D-PAS and PAS.The pathology changes were also observed by hematoxylin and eosin stain.The expression of type Ⅰ and Ⅲ collagen in liver tissues were examined by immunohistochemisty.Results： The level of serum TC,TG and the amount of 24 urinary proteins greatly decreased in four treated groups compared with the model group（P0.05）.The glycogen accumulation and fibration were alleviated in the liver tissues of rats treated by CHYS,accompanying the suppression of type Ⅰ and Ⅲ collagen expression in the liver.Conclusion： CHYS has protective effect on experimental liver injuries in diabetic nephropathy rats.
