摘 要:目的:观察补肾、健脾、活血方法对骨质疏松症大鼠骨骼、骨骼肌Na+-K+-ATP酶mRNA表达的影响,探讨中医防治骨质疏松症的作用机制。方法:将120只雌雄各半的大鼠随机分为正常组、模型组、补肾组、健脾组、活血组和骨疏康组。采用RT-PCR方法测定大鼠骨骼及骨骼肌Na+-K+-ATP酶mRNA表达。结果:与正常组相比,模型组大鼠骨组织和骨骼肌组织Na+-K+-ATP酶mRNA表达水平明显下降(P<0.05,P<0.01);与模型组相比,补肾组和健脾组大鼠的骨组织和骨骼肌组织Na+-K+-ATP酶mRNA表达水平明显升高(P<0.01);活血组及骨疏康组骨组织和骨骼肌组织Na+-K+-ATP酶mRNA表达低于补肾组及健脾组(P<0.01,P<0.05)。结论:骨质疏松症的形成可能与骨骼及骨骼肌Na+-K+-ATP酶表达异常有关;补肾、健脾方法通过调控骨质疏松症大鼠的骨骼及骨骼肌Na+-K+-ATP酶mRNA表达,对骨质疏松症具有一定的防治作用。Objective: To observe the changes of mRNA expression of Na+-K+-ATPase in the skeleton and skeletal muscle of osteoporosis rats and explore the effective mechanism of TCM on osteoporosis.Methods: 120 rats(male/female=1:1) was randomly divided into 6 groups as following blank group,model group,nourishing kidney group,invigorating spleen group,promoting circulation group and Gusukang group.RT-PCR method was used to test the expression of Na+-K+-ATPase in the skeleton and skeletal muscle.Results: Compared with normal group,mRNA expression of Na+-K+-ATPase in model group was significantly low(P0.05,P0.01);compared with model group,mRNA expression of Na+-K+-ATPase in treatment groups increased,among which nouring kidney group and invigorating spleen groups showed the greatest difference(P0.01),Compared with nourishing kidney group and invigorating spleen group,promoting circulation group and Gushukang group showed significant difference(P0.05,P0.01).Conclusion: The expression of Na+-K+-ATPase in the skeleton and skeletal muscle might play an essential role in the procedure of osteoporosis forming.Treatment of nourishing kidney and invigorating spleen showed certain treating and preventing effects on osteoporosis by regulating the expression of mRNA expression of Na+-K+-ATPase.
