云南琵琶甲提取物抑制HeLa细胞增殖和诱导凋亡的研究

摘　　要：目的:研究云南琵琶甲粗提物对人子宫颈癌HeLa细胞的增殖抑制作用,并进一步探讨其提取物抑制肿瘤细胞增殖的机制。方法:用水提反复冻融法得到云南琵琶甲提取物,倒置显微镜观察、MTT法检测琵琶甲提取物对HeLa细胞的抑制作用,荧光染色观察、流式细胞术及琼脂糖凝胶电泳法检测药物诱导细胞凋亡的作用。结果:云南琵琶甲粗提物对HeLa细胞有明显的增殖抑制作用,IC50为413μg/mL,荧光染色可见细胞皱缩,染色质凝集并边缘化,琼脂糖凝胶电泳可以看到梯状条带,流式细胞术发现S期细胞增多。结论:确定云南琵琶甲提取物可抑制HeLa细胞增殖,并初步推测云南琵琶甲粗提物能够诱导HeLa细胞凋亡。Objective： To study the effects of proliferation inhibition and the apoptosis induction of the Balaps japanensis yunnanensis Mars.extracts on HeLa cells and explore further its mechanism.Methods： Yunnan Balaps japanensis yunnanensi extract was gotten using multigelation method.MTT method was used to assay the cell proliferation inhibition rate;the cell morphological changes were observed with microscope;the cell apoptosis was assayed by DNA Ladder method.Flow Cytometry was used to detect the effects of the extracts on cell cycle.Results： Balaps japanensis yunnanensis Mars.extracts significantly inhibited HeLa cell proliferation.The IC50 was 413μg/mL.The cells treated with the extracts showed chromatin compaction margination and nuclear fragmentation.A characteristic DNA ＂ladder＂ was shown on the agarose gel.Most of the cells were blocked in S stage.Conclusion： The extracts of the Balaps japanensis yunnanensi Mars.inhibited the HeLa cell proliferation and one of the reason speculated was inducing the cell apoptosis.
