摘 要:目的探讨视网膜色素上皮(retinal pigment epithelial,RPE)细胞表面是否有CD157的表达及其对RPE细胞黏附和迁移的作用。方法分别采用免疫组织化学、West-ern-blot、RT-PCR验证RPE细胞表面有无CD157的表达。取500×103个RPE细胞用10mg.L-1CD157抗体处理20min后种植于8μm孔径的Tanswell小室,置37℃、含体积分数5%CO2培养箱孵育18h检测细胞迁移变化;100×103个细胞用10mg.L-1CD157抗体处理20min后种植于10mg.L-1FN包被的Tanswell小室,37℃、含体积分数5%CO2培养箱孵育1h检测细胞黏附能力变化。迁移和黏附实验均用β-actin鼠抗作对照。结果免疫组织化学、Western-blot及RT-PCR均证实RPE细胞有CD157的表达。与对照组相比,CD157抗体刺激RPE细胞后,细胞的迁移(实验组迁移细胞数:203±36、219±42、225±37;对照组迁移细胞数:384±46、357±38、362±57)和黏附(实验组黏附细胞数:222±20、243±34、237±26;对照组黏附细胞数:330±56、353±44、347±38)能力均下降,差异均有统计学意义(均为P<0.05)。结论 RPE细胞表面可表达CD157,其与配体结合后对RPE细胞的迁移和黏附起抑制作用。Objective To investigated the expression of CD157 in retinal pigment epithelial(RPE) cells and its effects on RPE cells adhesion and migration. Methods Immunohistochemical, Western-blot and RT-PCR were utilized to detect the expression of CD157 in RPE cells. A total of 50 x 103 cells were pretreated for 20 minutes by 10 mg · L-1 CD157 antibody and then planted into 8 μm aperture Tanswell unit,incuba- ted in 37 ℃ 5% CO2 incubator for 18 hours,the RPE cells migration was observed. A to- tal of 100 × 103 cells were pretreated for 20 minutes by 10 mg · L- 1 CD157 antibody and then planted into FN-coated Tanswell unit,incubated 1 hour in 37 ℃ 5% CO2 incubator to detect adhesion ability. Anti-murine β-actin was set as control antibody in migration and adhesion experiments. Results Immunohistochemical, Western-blot and RT-PCR techniques confirmed the expression of CD157 on RPE cells. Compared with the control group,average migration( experimental group cells:203± 36,219 ± 42,237 ± 26;Control group cells: 384 ± 46,353 ± 44,362 ± 57 ) and adhesion ( experimental group cells: 222 ± 20,243 ± 34,237 ± 26 ; Control group cells: 330± 56,353 ± 44,347 ± 38 ) abilities of the RPE cells pretreated with CD157 antibody decreased, there were statistical differences ( both P 〈 0.05 ). Conelusion RPE cells express CD157, which can inhibit the migration and adhesion of RPE cells after binding to the ligand.
