肾疏宁对体外培养大鼠腹膜间皮细胞AQP1表达的影响

摘　　要：目的:探讨肾疏宁对体外培养大鼠腹膜间皮细胞水通道蛋白1表达的影响。方法:原代培养大鼠腹膜间皮细胞、鉴定后传至第3代后分为5组:空白血清组、1.5%腹膜透析液组、4.25%腹膜透析液组、1.5%腹膜透析液+肾疏宁含药血清组、4.25%腹膜透析液+肾疏宁含药血清组。分别采用ELISA酶联免疫法、荧光定量PCR检测水通道蛋白1的表达。结果:1.5%PDS组、4.25%PDS组、1.5PDS%+肾疏宁含药血清组、4.25%PDS+肾疏宁含药血清组各组均与空白血清组比较AQP1表达指数均增高(P<0.05)。结论:证明肾疏宁含药血清有上调体外培养大鼠腹膜间皮细胞上水通道蛋白1的表达的作用。Objective:To explore the role of shengshuning in the protein expression of Aquaporin 1 harvested from rat peritoneal mesothelium cells in vitro.Methods:Cultured and identified rat peritoneal mesothelium cells in vitro.Then the third generations were divided into 5 groups:blank serum group,1.5% peritoneal dialyzate group,4.25% peritoneal dialyzate group,1.5% peritoneal dialyzate combined with shenshuning medicated serum group,4.25% peritoneal dialyzate combine with shenshuning medicated serum group.The expression of Aquaporin 1 was investigated respectively by means of Enzyme-linked immunosorbent assay and fluorescence quantitative PCR.Results:The AQP1 expression in the 1.5% peritoneal dialyzate group,4.25% peritoneal dialyzate group,1.5% peritoneal dialyzate combined with shenshuning medicated serum group,4.25% peritoneal dialyzate combined with shenshuning medicated serum group is significantly higher than the blank serum group(P<0.05).Conclusion:Shenshuning medicated serum increases the expression of aquaporin 1 in rat peritoneal mesothelium cells in vitro.
