摘 要:目的调查新疆部分地区人群及动物感染恙虫病东方体(Ot)情况,并对媒介昆虫进行调查。方法采用多种方法捕获可能感染Ot动物取脾,当地牧民静脉取血,从野鼠体表捕获恙螨,用试剂盒提取DNA,应用巢式PCR(nPCR)检测Ot-Sta56基因;接种昆明小白鼠进行Ot病原传代分离。结果共采取人群血液2430份,在6份血液中检测并分离到Ot;捕获动物13种5783个(只),从5种啮齿动物(小家鼠、灰仓鼠、大砂土鼠、草原兔尾鼠、灰旱獭)、2种鸟类(麻雀、灰腹鹡鸰)的脾中检测并分离到Ot,从绵羊血液中检测到Ot。其他动物体内未检测到Ot。从野鼠体表分离恙螨5种,从博乐纤恙螨体内检测分离到Ot。基因序列分析表明,新疆地区存在恙虫病东方体的基因型以Karp型为主,可能存在Saitama型。结论新疆地区存在人群及动物感染Ot,其基因型以Karp为主,可能存在Saitama型。Objective To investigate the infection in mankind and animal of Orientia tsutsugamushi in Xinjiang, China. Methods DNAs were extracted from spleens of the captured mice and the blood of the worker and the Chigger mite were seized from the rodent body surface. Nested polymerase chain reaction (nPCR) technique was used to detect the Ot Sta 56 gene. Some positive samples were sequenced and analyzed by BLAST. Results We detected the positive results from the pasturing worker, rodents (M.musculus, C.migratorius, R.opimus, L.luteus, M.baibacina), birds(L.passer domesticus, T.motacilla cinerea) and sheep. Orientia tsutsugamushi were isolated from the rodent spleen and Chigger mite. The Sta56 gene nucleotide sequence was homology to Karp strain. The Saitama strain could exist in certain areas. Conclusion The infection of Orientia tsutsugamushi in mankind and animal existed in Xinjiang.
