摘  要:目的从氧化应激角度探讨细颗粒物(PM2.5)对人脐静脉内皮细胞的影响,并探讨人参皂甙Rgl(简称Rgl)对其的干预效果。方法以人脐静脉内皮细胞为研究对象,筛选PM2.5合适的染毒浓度和Rgl的最适浓度。首先设对照组和PM2.5处理组(浓度分别为0.1、0.2、0.4、0.8mg/m1)。再用Rgl进行干预,设对照组、PM2.5,0.8mg/ml组、PM2.5,0.8mg/ml+Rg10.04mg/ml组和Rg10.04mg,/ml组。各组处理24h后用MTY法测定内皮细胞生长变化的吸光光度值(A值),荧光探针标记法测定内皮细胞活性氧(ROS)水平,RT-PCR法测定血红素氧合酶-1(HO.1)、核因子E2相关因子2(Nrf2)mRNA表达水平。结果Rgl的最适浓度为0.04mg/ml。PM2.5,各处理组MTT法测定的A值均低于对照组,PM2,0.4和0.8ms/ml组与对照组比较P分别〈0.05和〈0.01。PM2.5各处理组ROS荧光灰度值均高于对照组(P均〈0.01)。PM2.5,各处理组HO-1平均A值均高于对照组,PM2.5,0.8mg/ml组与对照组比较P〈0.01。PM2,各处理组Nrf2平均A值与对照组比较差异均元统计学意义。PM250.8mg/ml十Rg10.04mg/ml组细胞』4值高于PM2.5 0.8mg/ml组(P〈0.01),ROS荧光灰度值低于PM2.50.8mg/ml组(P〈0.01),HO-1平均A值高于PM250.8mg/ml组(P〈0.05),Nrf2平均A值与对照组比较差异无统计学意义。结论氧化应激是PM2.5,损伤内皮细胞的途径之一。Rgl能抑制PM2.5,损伤内皮细胞,抗氧化应激是其机制之一。但在Nrf2-ARE抗氧化通路中Nrf2对抗氧化蛋白HO-1表达的调节发生在其转录后水平。Objective To explore the mechanism of fine particulate matter (PM2.5) induced endothelial injury and the efficacy and mechanism of ginsenoside Rgl on the inhibition of endothelium injuries in human endothelial ceils exposured to PM2.5 Methods Human umbilical vein endothelial cells (HUVECs) were stimulated with various concentrations PM2.5 (0. 1, 0. 2, 0. 4, 0. 8 mg/ml) and PM2.5 at concentration 0. 8 mg/ml induced significant endothelial injury and was chosen for the main study in the presence or absence of Rgl (0. 04 mg/ml). After 24 h treatment, cell growth A value was detected through MT'F, intracellular reactive oxygen species (ROS) level through fluorescence labeling probe method and HO-1, Nrt2 mRNA expression was detected by RT-PCR. Results The cell A value was significantly lower while the ROS fluorescence gray value and the average optical density ratio of HO-1 were significantly higher in PM2.5 group than in the control group ( all P 〈 0. 05 ). The average optical density ratio of Nrt2 was similar between PM2.5 group and control group ( P 〉 0. 05). The A value and the average optical density ratio of HO-1 were significantly higher while the ROS fluorescence gray value was significantly lower in co-treated PM2.5 ( 0. 8 rag/m1) + Rgl (0. 04 mg/ml) group than in the PM2.5 (0. 8 rng/ml) group (all P 〈0. 05). Conclusion PM2.5 could induce human endothelial ceils injury by increasing oxidative stress which could be attenuated by ginsenoside Rgl.
