胸腺内注射供体MHC抗原对同种异体肢体移植的影响

摘　　要：目的探讨胸腺内注射异基因抗原对同种异体肢体移植存活的影响。方法 Wistar大鼠为供体,SD大鼠为受体。将SD大鼠随机分成3组,A组:自体肢体移植组;B组:异体肢体移植组;C组:胸腺内注射组。术后观察肢体的存活时间,并在术后7 d进行移植肢体皮肤苏木精-伊红染色检查及血清IFN-γ细胞因子的检测。结果 C组肢体存活时间长于B组,差异具有统计学意义(P<0.05)。组织学检查发现,C组出现的炎症排斥反应以及组织的改变明显较B组轻,与A组相接近。IFN-γ的检测,C组低于B组,差异具有统计学意义(P<0.05)。结论胸腺内注射异基因抗原可以对异体肢体移植产生特异性免疫耐受作用。Objective To investigate effect of intrathymic injection of allogeneic antigen on limb allograft survival.Methods Wistar rats were used as donors,and SD rats were used as receptors.The SD rats were randomly divided into 3 groups： Group A（autologous limb transplantation group）,group B（limb allograft group）,and group C（intrathymic injection group）.Limb survival time after operation was recorded.Seven days after operation,hematoxylin-eosin staining and serum IFN-γ concentration were detected.Results Limb survival time was significantly longer in group C compared with that in group B（P0.05）.Histological analysis showed that the rats in group C had less inflammation and histological changes compared with those in group B.There was no significant difference between group C and group A.The serum IFN-γ level was statistically lower in group C compared with that in group B（P0.05）.Conclusion The intrathymic injection of allogeneic antigen on limb allograft can generate specific immune tolerance.
