摘 要:目的:探讨体外培养类风湿关节炎(RA)患者滑膜成纤维细胞对T淋巴细胞生物学行为的影响。方法:滑膜组织取自关节置换术的RA患者,酶消化法获得滑膜成纤维细胞并扩增培养,T淋巴细胞选用Jurkat T淋巴细胞株。将T淋巴细胞按培养方式分为单纯T淋巴细胞培养组及与RA滑膜成纤维细胞共育培养组2组。分别采用噻唑蓝(MTT)法、流式细胞仪和实时荧光定量PCR方法检测2组T淋巴细胞的扩增能力、凋亡率及白细胞介素(IL)-2 mRNA、IL-23p19 mRNA的表达水平。结果:共育培养组4d和7d时T淋巴细胞增殖率高于单纯培养组,而10d和13d时增殖率低于单纯培养组,差异均有统计学意义(P<0.01);共育培养组24h和48hT淋巴细胞凋亡率均低于单纯培养组,差异有统计学意义(P<0.05);共育培养组较单纯培养组T淋巴细胞表达较高的白细胞介素(IL)-2 mRNA(2.33±0.09vs1.00±0.12)和IL-23p19 mRNA(4.01±0.25vs1.00±0.15),差异均有统计学意义(t分别为15.03、17.78均P<0.01)。结论:RA滑膜成纤维细胞可诱导T淋巴细胞的激活,这可能是RA发病及进展的影响因素之一。Objective: To investigate the effect of rheumatoid synovial fthroblast on biological properties of T cells. Methods: The sample of synovial tissue was obtained from rheumatoid arthritis (RA) patients with joint replacement surgery. The synovial fibroblasts were extracted by enzyme. The Jurkat T cell line was used in the experiment. These T cells were divided into two groups, T cell alone group and co-culture group according to whether cells were co-cultured with RA fibroblast. MTT, flow cytometry and real-time PCR were used to detect the proliferation ability, the apoptosis and the expression of interleukin (IL)-2 mRNA and IL-23p19 mRNA of T cells, respectively. Results: The proliferation rate was significantly higher in co-cuhure group than that ofT cell alone group at the 4^th d and the 7^th d respectively (P 〈 0.01), which was significantly lower in co-euhure group than that of T cell alone group at the 10^thd and the 13^thd respectively (P 〈 0.01). The apoptotic rate was significantly lower in co-culture group than that of T cell alone group at the 24^thh and the 48^thh respectively (P 〈 0.05). The expression levels of IL-2 mRNA (2.33±0.09 vs 1.00±0.12) and IL-23p19 mRNA (4.01±0.25 vs 1.00±0.15) were significantly higher in co-culture group than those in T cell alone group (P 〈 0.01). Conclusion: RA fibroblasts have the potential effect on activating T cells, which may be one of factors involved in RA pathogenesis and progression.
