摘  要:目的:观察石斛合剂对衰老及衰老糖尿病大鼠胰腺组织凋亡相关基因P53表达的影响。方法:实验分为正常对照组、衰老模型组、衰老糖尿病模型组、石斛合剂组。除正常组外,大鼠腹腔注射D-半乳糖致衰老,继以高脂高糖加腹腔注射链脲佐菌素造成衰老糖尿病。疗程结束后测禁食血糖,及利用RT-PCR及Western-blot法检测胰腺凋亡相关基因P53 mRNA及蛋白表达水平。结果:与正常对照组相比,衰老模型组大鼠禁食血糖正常(P>0.05),但衰老糖尿病及治疗组大鼠血糖均升高(P<0.05),衰老糖尿病模型组大鼠胰腺组织P53 mR-NA及蛋白水平明显升高(P<0.01~0.05);与衰老糖尿病模型组相比,石斛合剂组禁食血糖下降(P<0.05),胰腺组织P53 mRNA及蛋白水平明显降低(P<0.01)。结论:衰老糖尿病模型大鼠呈现出血糖升高,胰腺组织细胞凋亡增多;而石斛合剂可能通过抑制胰腺组织细胞的凋亡而达到降血糖的作用。Objective : To study the influence of Dendrobium Compound ( DC ) on mRNA and protein expression of P53 gene associated with apoptosis in aged diabetic rats pancreas. Methods : The aged diabetic rat model was induced by D - galactose, enrich - diet and Streptozotocin to measure the concentration of blood glucose in serum, and detect mRNA and protein expressions of P53 gene by RT - PCR and Western - Blot. Results : Compared to the control group, the level of blood glucose in aged group was unchanged(P 〉0.05 ) ,those of aged diabetic and DC treat groups were heightened(P 〈 0. 05 ),and P53 mRNA and protein expression in aged and aged diabetic groups were evidently inclined( P 〈 0.01 0. 05 ) ; compared to the aged diabetic group, the level of blood glucose in DC treated group was declined (P 〈 0.05 ) , and P53 mRNA and protein expression in DC treated group were also reduced ( P 〈0. 05 ). Conclusions : It demonstrated that the blood glucose raised and pancreatic cell apoptosis increased in the aged diabetic group, and DC could lower blood glu- cose via inhibiting the pancreatic cell apoptosis.
