摘 要:目的:观察补肾活血方补肾活血方对前列腺增生大鼠膀胱逼尿肌缝隙连接蛋白43(Cx43)mRNA及蛋白表达的影响。材料与方法:清洁级雄性Wistar大鼠48只,随机分为正常组、模型组、模型加补肾活血方组、模型加坦索罗辛组四组。除正常组外,其它实验大鼠以去势加睾酮注射法复制大鼠BPH模型。于去势第一天起,各组大鼠分别给予相应药物灌胃。模型+补肾活血方按1mL/100g/日体重予补肾活血方(由淫羊藿、枸杞子、穿山甲、丹参、菟丝子等组成,含生药量2.34g/ml)灌胃,模型加坦索罗辛组按1.8μg/100g/日体重予坦索罗辛灌胃。其余各组予等体积生理盐水灌胃。均每日1次,连续38天后检测各组大鼠膀胱逼尿肌组织Cx43 mRNA及蛋白表达。结果:与空白组相比,模型组Cx43 mRNA表达明显上升(2.08 vs 30.69,P<0.01),坦索罗辛及补肾活血方组Cx43 mRNA表达与空白组相比也有升高,但无统计学差异(16.05,12.74,2.08,P>0.05)。与模型组相比,补肾活血方(12.74 vs 30.69,P<0.01)Cx43 mRNA表达明显下降。与空白组相比,模型组及补肾活血方、坦索罗辛等各组Cx43蛋白表达明显上升(分别为1.44,3.89,1.86,2.73),而补肾活血方、坦索罗辛各组与模型组相比,Cx43蛋白表达均明显下降(P<0.01)。结论:补肾活血方组可以明显降低BPH模型大鼠膀胱组织Cx43 mRNA及蛋白表达,这可能是补肾活血方有效改善BPH继发膀胱改变的机制之一。Objective:To observe the effect of Bushen Huoxue Formula on mRNA/protein expression of connexin 45 ( Cxd3 ) in the detrusor of benign prostatic hyperplasia( BPH ) rats. Methods : Clean grade 48 male Wistar rats were random- ly divided into four groups(A -D group). Except A group,the other groups received castration plus estosterone injection to make model of BPH. On the first day of castration,the rats were given the appropriate medication orally. C group ga- raged with Bushen Huoxue Formula(1mL/( 100g · day), including herbs with 2.34g/mL), D group received Tamslusin with 1.8μg/( 100g · day) ,and A and B groups received the same volume saline. The mRNA/protein expression of Cx43 in the detrusor tissue of all groups was detected after 38 days. Results :Compared with the control group,the Cx43 mRNA expression of B group was significantly increased(2.08 vs 30.69 ,P 〈 0.01 ). The Cxd3 mRNA expression also increased in C and D groups compared with the blank group ,but without statistical difference( 16.05,12.74,2.08, P 〉 0.05 ). Com- pared with the B group,Cx43 mRNA expression was significantly decreased in C group( 12.74 vs 30.69 ,P 〈0.01 ). The protein expression of Cx43 was significantly increased in B, C, D groups compared with that in the A group(3.89,1.86, 2.73 respectively vs 1.44 ). But protein expression of Cx43 was significantly decreased( P 〈 0.01 ) compared with that inthe B group. Conclusion : Bushen Huoxue Formula can significantly downregulate the mRNA/protein expression of connexin 43 (Cx43)in the detrusor of benign prostatic hyperplasia rats, which may be one of the mechanisms of Bushen Huoxue Formula protecting bladder function of BPH rats.
