摘 要:目的评价VEGF同轴静电纺丝膜的物理特性、体外血液相容性和生物学活性,探求其作为人工心血管替代材料的可行性。方法通过静电纺丝技术制备VEGF同轴静电纺丝膜,然后进行体外溶血实验、动态凝血实验、凝血时间测定、血小板粘附实验、细胞毒性实验、体外细胞粘附实验对其血液相容性和生物学活性进行评价,并与涤纶(Dacron)作平行对照。结果 VEGF同轴静电纺丝膜的血液相容性较高,经统计学分析与Dacron之间差异无统计学意义(P>0.05)。VEGF同轴静电纺丝膜的体外细胞粘附实验中,经细胞计数Dacron为(5.34±0.94)×103,PCL-PEG静电纺丝膜为(7.82±0.85)×104,PCL-PEG/BSA同轴静电纺丝膜为(8.04±0.84)×104,PCL-PEG/BSA-VEGF同轴静电纺丝膜则为(1.39±0.76)×105,经统计学分析PCL-PEG静电纺丝膜和PCL-PEG/BSA同轴静电纺丝膜之间差异无统计学意义(P>0.05),PCL-PEG静电纺丝膜和PCL-PEG/BSA同轴静电纺丝膜与Dacron之间差异有统计学意义(P<0.05),而PCL-PEG/BSA-VEGF同轴静电纺丝膜与PCL-PEG静电纺丝膜和PCL-PEG/BSA同轴静电纺丝膜之间的差异具有统计学意义(P<0.05)。结论 VEGF同轴静电纺丝膜血液相容性较高,且更易于与内皮细胞粘附,显示其成为人工心血管替代材料的前景。Objective To evaluate the blood compatibility,physical characteristics and biological activity of the VEGF-coaxial electrospinning membrane in vitro and explore its feasibility to be used as substitute of artificial cardiovascular materials.Methods The VEGF-coaxial electrospinning membrane was prepared by coaxial electrospinning technique.The blood compatibility of the VEGF-coaxial electrospinning membrane was investigated in terms of hemolysis,dynamic clotting time,thrombin time(TT),prothrombin time(PT),activated partial thromboplastin time(APTT),platelet adhesion,cytotoxicity test and biological activity,and compared with Dacron.Results The VEGF-coaxial electrospinning membrane was highly compatible with human blood,which had no significant difference from Dacron(P0.05).In the cell adhesion test of the VEGF coaxial electrospinning membrane in vitro,the cell count in Dacron group,PCL-PEG group,PCL-PEG/BSA group and PCL-PEG/BSA-VEGF group was(5.34±0.94)×103,(7.82±0.85)×104,(8.04±0.84)×104,and(1.39±0.76)×105,respectively.There was no significant difference between PCL-PEG group and PCL-PEG/BSA group(P0.05),but significant difference was found between Dacron group and PCL-PEG group or PCL-PEG/BSA group(P0.05),and between PCL-PEG/BSA-VEGF group and PCL-PEG group or PCL-PEG/BSA group(P0.05).Conclusion The VEGF-coaxial electrospinning membrane is highly compatible with human blood and is apt to be adhered by vascular endothelial cells,so it shows favorable prospects to be as substitute of artificial cardiovascular materials.
