摘  要:目的:探讨枸杞多糖对人肝癌细胞Bel-7402凋亡及其周期的影响。方法:利用MTT染色在普通光镜下观察细胞凋亡形态学变化并观测枸杞多糖对肿瘤细胞的抑制率;通过流式细胞仪分析人肝癌细胞Bel-7402凋亡率、细胞周期变化。结果:经枸杞多糖作用后的人肝癌细胞Bel-7402出现核固缩、凋亡小体、新月状浓染区等细胞凋亡形态学改变,各剂量LBP组的肿瘤细胞均表现生长抑制,且具有剂量依赖性,1000μg/ml LBP处理组的抑制率达96.02%。药物处理组的肿瘤细胞更多地被阻止于G2/M期,各剂量组均出现明显的凋亡变化,最大凋亡率可达21.3%。结论:枸杞多糖可明显抑制人肝癌细胞Bel-7402的生长增殖,诱导细胞凋亡,并能改变该肿瘤细胞周期分布,阻止细胞周期于G2/M期。Objective The influnce of lycium bararum polysaccharides (LBP) on apoptosis and the cell cycle of Human hepatocarcinoma cell Bel-7402 were studied. Methods The cell morphology of apoptosis and inhibitory rate of the cancer cell were analyzedby MTT staining and a ordinary microscope. The apoptosis rate of cells Bel-7402 and cell cycles were studied by the flow cytometer. Results After treatment with the LBP, Be1-7402cells showed morphologic features of apoptosis such as pyknosis, apoptotic body, thick stained area as crescents.Treated cancer cells were inhibited and dose dependent and maximal inhibition rate was 96.02%. After treatment with LBP,most Bel-7402 cells were arrested in the G2/M phase and appeared apoptosis, maximal apoptosis rate of Bel-7402 was 21.3%. Conclusion LBP can obviously inhibit human esophageal carcinoma cell Bel-7402 to proliferate, induce the apoptosis and arrest the cell cycle in the G2/M phase.
