摘 要:目的:研究香丹注射液对兔骨性关节炎(OA)的疗效及其作用机制。方法:40只新西兰大白兔,随机分组为5组,阴性对照组(A组),阳性对照组(B组),给药治疗低剂量组(C组),给药治疗中剂量组(D组),给药治疗高剂量(E),每组8只。A组不造模,B、C、D、E组均采用Hulth法建立OA模型,术后3天给予生理盐水,C、D、E组给予香丹注射液,C组,每天0.05mL/100g;D组,每天0.1mL/100g;E组,每天0.2mL/100g。自术后1周每天驱赶动物2次,共30分钟。4周后处死动物,对组织结构进行观察并制备待测样品。采用免疫组化监测法分别检测关节软骨和滑膜中的白介素-1β(1L-1β)。采用硝酸还原酶法测定血清和关节液的NO含量,动物处死后对软骨及滑膜组织的匀浆进行iNOS的测定。结果:B组1L-1β的水平显著高于A组第一作者杨福彪(p<0.01),并且高于C、D、E组(0.01<p<0.05)。C、D、E组的关节液NO和iNOS含量降低的程度与B组比较明显,但是均明显低于B(p<0.05)。结论:香丹注射液能有效抑制1L-1β产生,通过抑制iNOS的表达,可降低NO含量。具有抗炎、保护和修复关节软骨的作用,从而对OA、滑膜炎有治疗作用。Objective: To study therapeutic effect and mechanism of Xiangdan injection treating osteoarthritis in rabbits. Methods: Forty New-Zealand white rabbits were divided into 5 groups: the 1 blank group (A group), the 4 model groups (B.C.D and E group). Every group was 8 rabbits. A group was not taken any measures. B, C, D and E groups were established OA model with Hulth-method. At the fourth day after surgery, all groups are given saline. The B group is in injected with saline. The C.D and E groups are injected with the low, middle and high dosage of Xiangdan injection (0.05mL/100g, 0.1 mL/100 g and 0.2 mL/100 g per day). At the second week after surgery, rabbits need to be forced to move 30 mins per day. After 4 weeks, all animals were killed. Observe the organizational structure of knee and prepare the sample under test. The synovium and cartilage which all groups were treated were tested the concentrations of 1L-1 and iNOS level. The concentrations of nitrogen monoxide (NO) of the serum and joint fluid were tested. Results: The 1L-1βand iNOS level of the synovium and cartilage in Xiangdan injection (C, D, E groups) and blank group (A group) were lower than model group (B group) .and there was significantly differently. The NO concentration of the serum and joint fluid was the same trend as the 1L-1β and iNOS level. Conclusions: Xiangdan injection can reduce the concentration of NO and 1L-1β. It has anti-inflammatory and protects the cartilage effect. Xiangdan injection has therapeutic effects to OA and synovitis.
