摘  要:通过点眼将IBDV感染4周龄SPF鸡,采用相对荧光定量PCR技术检测SPF鸡腔上囊单核细胞(BBMC)和外周血单核细胞(PBMC)在感染后不同阶段TLR3/4/15mRNA表达动态变化,探讨IBDV感染对免疫反应的影响。结果显示,感染后1dBBMC中TLR3、15的mRNA表达水平高于对照组,差异显著(P≤0.05);在感染后5dTLR3、4显著高于对照组(P≤0.05);在感染后15dTLR3/4/15的mRNA表达水平恢复到正常水平。PBMC中,感染后1dTLR3、4mRNA表达水平均显著高于对照组(P≤0.05),TLR15极显著高于对照组(P≤0.01);而在感染后5d和15d,TLR3/4均显著高于对照组(P≤0.05)。以上结果表明,IBDV感染早期上调了腔上囊和外周血单核细胞的TLR3/4/15的mRNA表达水平,增强了机体的固有免疫反应。Four-week-old SPF chicks were infected with infectious bursa disease virus (IBDV) by eye drops. To investigate host immune response to IBDV infection, the transcription levels for TLR3, TLR4 and TLR15 in bursa mononuclear cells (BBMC) and peripheral blood mononuclear cells (PBMC) of SPF chicks at different days post inoculation (dpi) were examined by relative quantitative PCR. Results showed that the transcription levels for TLR3/15 in BBMC were significantly up-regulated (P≤0.05) at 1 dpi. The expressions of TLR3/4 were up significantly (P≤0. 05) at 5 dpi, while there was no significant difference for TLR15. At 15 dpi, the levels of TLR3/4/15 were rebound. Compared to the control group, the transcrip- tion levels for TLR3/4/15 in PBMC were significantly higher (P≤0.05) at 1 dpi, 5 dpi, and 15 dpi(P≤ 0. 05) except TLR15 at 5 dpi and 15 dpi. All these data suggested that IBDV could increase the mRNA ex- pression of TLR3/4/15 in BBMC and PBMC at the early stages of infection, indicating that the natural im- mune response was enhanced.
