摘 要:目的研究醛固酮(ALD)对体外培养的肾小管上皮细胞凋亡的影响及其机制。方法人肾小管上皮细胞(HK-2)培养于无血清DMEM/F12培养液中,以不同浓度的ALD(0、10-11、10-10、10-9和10-8mol/L)刺激细胞48 h后,Annexin V/PI双染流式细胞术检测ALD对HK-2细胞凋亡的影响;Western blotting检测ALD对HK-2细胞中Bax和Bcl-2蛋白表达的影响。结果流式细胞术结果表明,随着ALD浓度的升高,凋亡细胞百分率逐渐增多,与对照组比较差异有统计学意义(P<0.05)。Western blotting结果表明,随着ALD浓度的升高,Bax蛋白表达量逐渐增加,Bcl-2蛋白表达量逐渐减少,Bax/Bcl-2比值增加。结论 ALD可以通过调节凋亡调控蛋白Bax和Bcl-2的表达促进肾小管上皮细胞凋亡,进而促进肾小管间质纤维化的进展。【Objective】 To investigate the effect of aldosterone(ALD) on apoptosis in cultured renal tubular epithelial cells and its mechanism.【Methods】 Human renal tubular epithelial cells(HK-2) were cultured in DMEM / F12 medium without FCS.Cells were exposed to different concentrations of ALD(0,10-11,10-10,10-9 and 10-8 mol/L) for 48 h.The apoptosis of HK-2 was detected by flow cytometry via Annexin V/PI staining.Western blot was used to measure the Bax and Bcl-2 protein expressions.【Results】 Flow cytometry results showed ALD increased significantly the rate of apoptotic HK-2 in a dose dependent manner,compared with control group(P 0.05).Western blot methods demonstrated the Bax protein expressions were up-regulated,and the Bcl-2 expressions were down-regulated in a dose dependent manner,and the ratio of Bax/Bcl-2 was increased apparently when HK-2 were treated with ALD.【Conclusions】 ALD can promote apoptosis of renal tubular epithelial cells by regulating Bax and Bcl-2 expressions,thus contributing to tubulointerstitial fibrosis.
