摘  要:目的对在临床治疗上有较好疗效的柴芪汤进行实验研究,探讨其干预代谢综合征(MS)的作用机制。方法采用高脂高糖高盐饮食喂养方法制备大鼠代谢综合征模型,并分为模型组、中药组、罗格列酮组和正常组。中药组从造模开始即给予柴芪汤,给药量为5.71 g/(kg.d);罗格列酮组从造模开始给予罗格列酮1.2 mg/(kg.d)。给药剂量按大鼠的体表面积进行计算。观察并比较不同组别大鼠的一般情况、实验室指标及肝脏组织学变化。用生化法检测丙氨酸氨基转移酶(ALT)、甘油三脂(TG)、空腹血糖(FBG),放免法检测空腹胰岛素(FINS)、硫代巴比妥酸法测定肝匀浆苯二醛(MDA)、黄嘌呤氧化酶法测定肝匀浆超氧化物歧化酶(SOD)、HE染色法观察肝脏的病理组织学改变。结果模型组大鼠一般情况逐渐变差,第8周代谢综合征造模成功。中药组及罗格列酮组肝功能、血脂、血糖、胰岛素敏感性、肝组织病理形态学改变及SOD活性、MDA含量等指标均优于模型组。中药组在改善TG、降糖及改善胰岛素敏感性方面稍逊于罗格列酮组,余指标改善情况均优之,但2组比较无统计学差异。结论柴芪汤对代谢综合征和非酒精性脂肪肝(NAFLD)均有较好的防治作用,既可以降低血清中胰岛素含量,改善胰岛素抵抗(IR)的状态,又可以改善脂代谢,可能是治疗MS和NAFLD的作用机制之一。Objective To study Chaiqi Tang with better curative effect in clinic, and investigate its mechanism in intervening metabolic syndrome. Methods The rat model of metabolic syndrome was established by feeding high fat, high sugar and high salt diet, and then all rats were divided into model group, Chaiqi Tang group, rosiglitazone group and normal group. The Chaiqi Tang group was given Chaiqi Tang (5.71 g/ kg · d) from the beginning of modeling, and rosiglitazone group was given rosiglitazone (1.2 mg/ kg · d, calculated according to rat body surface ). The changes of general condition, laboratory indexes and histological changes of liver were observed and compared in rats of all groups. The levels of alamine aminotransferase (ALT), triglyceride (TG) , and fasting blood glucose (FBG) were detected by using biochemical method, the level of fasting insulin (FINS) was detected by using radioimmunoassay, the level of malondialdehyde (MDA) in liver homogenate was detected by using thiobarbituric acid method, the level of superoxide dismutase (SOD) in liver homogenate was detected by using xanthine oxidase method, and histopathological changes of liver were observed by using HEstaining. Results The general condition became worse in rats of model group, and the model of metabolic syndrome was successfully established on the 8th week. Chaiqi Tang group and rosiglitazone group were superior to model group in the improvement of indexes of liver function, blood fat, blood glucose, insulin sensitivity, liver histopathology, SOD activity and level of MDA. Chaiqi Tang group was marginally worse than rosiglitazone group in improving TG and insulin sensitivity and reducing blood glucose, and superior to in other indexes, but there was no significant difference between two groups. Conclusion Chaiqi Tang has a good therapeutic effect on metabolic syndrome and non-alcoholic fatty liver disease, and it can reduce the content of serum insulin, improve insulin resistance state as well as improve lipid meta
