摘 要:采用集合分离分析法(Bulked segregant analysis,BSA),结合SSR分子标记技术筛选与抗病基因Ty-2连锁的标记。将感病材料TMXA48-4-0与含有Ty-2的抗病材料CLN2777A杂交,获得F2分离群体。利用前人初步定位的结果及构建的番茄SSR遗传图谱,选择11号染色体上TG36与TG393标记之间的SSR标记对构建的抗感池进行筛选分析,结果得到1个与Ty-2紧密连锁的呈共显性的分子标记TES0344,遗传距离为6.7 cM。利用F2分离群体和10份同源材料进一步验证了SSR标记TES0344的可靠性和稳定性,因此该标记可用于今后抗番茄黄化曲叶病毒病育种的辅助选择。Tomato yellow leaf curl virus(TYLCV) is one of the most devastating viruses of cultivated tomatoes in the world,and the damage caused by TYLCV in China increased every year.Ty-2 is a gene highly resistant to the TYLCV in Jiangsu province and the surrounding regions.To find out the markers linked to Ty-2,bulked segregant analysis(BSA) and simple sequence repeat(SSR) molecular marker were used.The F2 segregation population derived from susceptible parent TMXA48-4-0 and resistant parent CLN2777A,and seven SSR markers that was located between TG36 and TG393 on the chromosome 11 of tomato,were used to detect molecular markers closely linked to gene Ty-2.One co-dominant marker TES0344 was identified with distances of 6.7 cM from Ty-2 gene.It was validated to be a reliable and stable marker in F2 population and 10 other resistance TYLCV accessions.These results suggested that TES0344 could be used in marker-assisted selection of TYLCV resistance in breeding of tomato.
