摘　　要：本文介绍表达序列片段法在筛选心血管病相关基因中的应用。从人主动脉文库中挑取有目的基因片段插入的噬菌斑 ,选用双向通用引物做PCR扩增 ,以纯化后的产物做测序模板 ,用全自动DNA测序仪测定基因表达序列并与GenBank作同源性分析。PCR扩增阳性克隆产率大于 70 % ,新ESTs占 15%。结果表明利用正常人主动脉cDNA文库 。Cardiovascular associated genes were screened with Express Sequence Tags (ESTs) technique in this report.Plaques were picked up from nomal human artery cDNA library and then PCR with vector primers was performed.And the purified PCR products were used for cycles sequencing.ESTs were obtained with DNA sequencer and they were compared with GenBank data base.Positive clone rate was more than 70% and about 15% of them were new ESTs.The results suggest that the technique of screening ESTs from human artery cDNA library is useful for screening cardiovascular associated genes.
