金莲花中荭草苷对人食管癌EC-109肿瘤细胞生长及凋亡的影响

摘　　要：目的观察金莲花中荭草苷对EC-109细胞体外生长增殖抑制以及诱导EC-109细胞凋亡的作用。方法用不同浓度荭草苷作用于对数生长期的EC-109细胞,通过CCK-8法检验其对EC-109细胞体外生长、增殖的抑制作用,通过凋亡试剂盒Hoechest33258荧光染色细胞形态观察、琼脂糖凝胶电泳检测DNA Ladder、AnnexinⅤ-FITC/PI双标法流式细胞术,观察荭草苷诱导EC-109细胞凋亡情况。结果荭草苷对EC-109细胞体外生长、增殖具有明显的抑制作用,能够诱导EC-109细胞凋亡,作用随着药物浓度的增高而增加、随着作用时间的延长而显著增高。结论金莲花中荭草苷可剂量依赖性抑制EC-109细胞生长增殖并可诱导肿瘤细胞凋亡。AIM To search the effects of orientin on cells growth and apoptosis of EC-109 cells.METHODS EC-109 cells in logarithmic growth phase were treated with different concentrations of orientin.The inhibiting effect on the growth in vitro and proliferation were detected by CCK-8 method.Cell nucleus were assayed by the cell death detection kit Hoechest33258,cell apoptosis was detected by DNA agarose gel electrophoresis,and the apoptosis rate of EC-109 cells was examined by using AnnexinⅤ-FITC/PI double-labled technique of flow cytometry.RESULTS Orientin had markedly inhibiting effect on proliferation of EC-109 cells.The inhibition ratio increased with the increase of the concentration and with the extension of reaction time.Orientin could induce apoptosis of EC109 cells.CONCLUSION Orientin can inhibit EC-109 cell growth in dose-dependent manner and induce apoptosis of tumor cells.
